Glyoxylate carboligase-based whole-cell biotransformation of formaldehyde into ethylene glycol via glycotaldehyde

Summary: A novel biocatalytic system was established for the synthesis of industrially relevant C2 chemicals, such as ethylene glycol, from formaldehyde. By improving the substrate access tunnel, the affinity and catalytic efficiency of the thermostable glyoxylate carboligase for formaldehyde were enhanced, leading to the production of ethylene glycol through the condensation of two molecules of formaldehyde into one molecule of glycolaldehyde. The study demonstrated a green approach for the biocatalytic synthesis of ethylene glycol from Cl compounds.

Constructing a methanol-dependent Bacillus subtilis by engineering the methanol metabolism

Summary: This study developed a synthetic methylotrophic Bacillus subtilis that can use methanol as a carbon source by adopting several strategies, including the heterologous expression of key enzymes, regulation at both the translational and transcriptional levels, stabilization of enzyme expression, and improvement of catalytic activity. The methanol consumption of the synthetic methylotrophic B. subtilis reached 4.09 g/L, demonstrating its potential for utilizing methanol efficiently. The strategies used in this study are generally applicable to other studies on synthetic methylotrophy.

Engineering a Highly Efficient Carboligase for Synthetic One-Carbon Metabolism

Summary: This study successfully converted OXC into GCS through enzyme mutation, enabling the direct condensation of two C1 units with high catalytic efficiency. The new variant MeOXC4 showed a catalytic efficiency in C1-C1 condensations more than 40 times higher than other enzymes, and was able to produce glycolate in vivo, offering new possibilities for synthetic C1 assimilation routes.

Variants of the FORCE pathways were evaluated through thermodynamic and stoichiometric analyses, showing promising results both in vitro and in vivo. This synthetic metabolic pathway demonstrates the potential for C1 compound utilization and integration with host metabolism for synthetic methylotrophy.
