Assessing evolutionary and developmental transcriptome dynamics in homologous cell types

Christian Feregrino et al.

Summary: Using single-cell RNA-sequencing data, the study examined the transcriptional dynamics of homologous cell types during embryonic patterning by detecting gene co-expression modules. The results revealed modules with high evolutionary turnover, while others seemed maintained to different degrees over developmental and evolutionary timescales.

Postnatal development in a marsupial model, the fat-tailed dunnart (Sminthopsis crassicaudata; Dasyuromorphia: Dasyuridae)

Laura E. Cook et al.

Summary: In their study, Cook, Pask, and colleagues focus on the ossification of the skull and forelimbs in the developing postnatal fat-tailed dunnart, highlighting its potential as a model organism for studying jaw and forelimb development in mammals. Marsupials possess unique biological features that offer valuable insights into various aspects of mammalian development, with their distinctive reproduction mode, altricial birth stage, and associated heterochrony setting them apart from placental and monotreme mammals.

Single-cell reconstruction with spatial context of migrating neural crest cells and their microenvironments during vertebrate head and neck formation

Jason A. Morrison et al.

Summary: By using label-free, unsorted single-cell RNA sequencing, this study identified novel markers of migrating neural crest cells and 266 invasion genes common to all BA1-4 streams. Perturbation analysis of a subset of invasion genes revealed their functional role in regulating neural crest cell behaviors. Comparison with other cell invasion phenomena showed a shared set of 45 genes, some of which demonstrated relevance to human neuroblastoma cell lines exposed to the in vivo chick embryonic neural crest microenvironment.

Ontogenetic origins of cranial convergence between the extinct marsupial thylacine and placental gray wolf

Axel H. Newton et al.

Summary: The study reveals that the skulls of the thylacine and wolf develop along nearly parallel growth trajectories, despite lineage-specific constraints and differences in the timing of ossification. The patterns of distinct cranial modularity and integration during development are unable to explain their adult convergence, suggesting a developmental origin for their convergent cranial morphologies through mosaic evolution within bone groups sharing common embryonic tissue origins. Homoplasy in gene regulatory networks associated with neural crest cells is also observed, indicating empirical links between adaptive phenotypic and genotypic convergence.

Mammalian face as an evolutionary novelty

Hiroki Higashiyama et al.

Summary: The anterior end of the mammalian face is characterized by a semi-mobile nose, covered ventrolaterally by the premaxilla, unlike other tetrapods where the upper jaw is the main feature. The premaxilla in therian mammals is a composite of the septomaxilla and the palatine remnant of the premaxilla in nontherian amniotes, indicating evolutionary differences. The mammalian-specific face is an evolutionary novelty, with the muzzle derived from the maxillary prominence and establishing a unique topographical framework.

Single-cell transcriptomic analysis of zebrafish cranial neural crest reveals spatiotemporal regulation of lineage decisions during development

David Tatarakis et al.

Summary: By conducting single-cell RNA sequencing of cranial NC cells in zebrafish during migration stages, the study reveals a transitional state before cell lineage specification, with the first decisions being skeletal and pigment lineages followed by neural and glial progenitors. Integration of data with a Wnt reporter line identifies gene cohorts with temporal responses and indicates a role for gene Atp6ap2 in melanocyte differentiation. The results show progressive emergence of cranial NC cell lineages and spatially restricted cell interactions regulating cell fate decisions.

Functional constraints during development limit jaw shape evolution in marsupials

Anne-Claire Fabre et al.

Summary: Marsupials exhibit differences in jaw shape influenced by reproductive mode and diet, showing convergence with placentals but lower evolution rate and disparity in mandible shape. Their specialized reproductive biology appears to strongly constrain the evolution of their jaws despite ecological and morphological diversity.

A single-cell atlas of human teeth

Pierfrancesco Pagella et al.

Summary: This study mapped the transcriptional landscape of various cell populations in human teeth at single-cell resolution, analyzing their stem cell populations and microenvironment in detail. The research revealed significant cellular heterogeneity in dental pulp and periodontium, with similar molecular signatures of stem cell populations but divergent microenvironments. The findings suggest that microenvironmental specificity could be a key factor in functional differences between highly similar stem cells, potentially influencing cell-based dental therapeutic approaches.
