Causes and consequences of baseline cerebral blood flow reductions in Alzheimer's disease

Oliver Bracko et al.

Summary: Baseline cerebral blood flow reductions are common in Alzheimer's disease, potentially causing cognitive impairment and brain pathology, with studies showing that increasing baseline CBF can improve performance in AD mouse models. However, there is still poor understanding of the molecular and cellular mechanisms causing these CBF deficits, and future research aimed at elucidating the interplay between different mechanisms and their role in AD patients could guide the development of therapies targeting CBF reductions.

Demonstration of age-related blood-brain barrier disruption and cerebromicrovascular rarefaction in mice by longitudinal intravital two-photon microscopy and optical coherence tomography

Adam Nyul-Toth et al.

Summary: Age-related blood-brain barrier disruption and cerebromicrovascular rarefaction play important roles in the pathogenesis of vascular cognitive impairment and dementia (VCID) and Alzheimer's disease (AD). Recent advances in geroscience research have provided new interventions for preventing these diseases. Sensitive and easy-to-adapt imaging methods are urgently needed for longitudinal assessment of changes in BBB permeability and brain capillarization in aged mice.

IGF1R signaling regulates astrocyte-mediated neurovascular coupling in mice: implications for brain aging

Stefano Tarantini et al.

Summary: This study found that disrupted IGF1R signaling pathway affects astrocyte responses to external stimuli, decreases the release of vasodilators, accelerates the metabolism and inactivation of EETs, leading to impaired neurovascular coupling.

Increased cognitive workload evokes greater neurovascular coupling responses in healthy young adults

Tamas Csipo et al.

Summary: Understanding how the brain allocates resources to meet the demands of active neurons is crucial. NVC, the hemodynamic response to increased metabolic demands of active brain regions, can be an early marker for cognitive impairment. Non-invasive NVC assessments can help detect early signs of cognitive impairment and dementia. Both fNIRS and TCD methods can differentiate NVC responses evoked by higher demand tasks.

Reversal of cerebral hypoperfusion: a novel therapeutic target for the treatment of AD/ADRD?

Fan Fan et al.

Summary: Alzheimer's disease and Alzheimer's disease-related dementias are global healthcare crises primarily affecting the elderly population, especially those with conditions like diabetes and hypertension. Despite current treatments being ineffective, a recent study suggests that reversing cerebral hypoperfusion may be a novel therapeutic target for these diseases, highlighting the importance of further research in this area.

Endothelial deficiency of insulin-like growth factor-1 receptor (IGF1R) impairs neurovascular coupling responses in mice, mimicking aspects of the brain aging phenotype

Stefano Tarantini et al.

Summary: The impairment of neurovascular coupling plays a role in the pathogenesis of vascular cognitive impairment, with endothelium-mediated microvascular dilation being central to NVC responses. Disruption of endothelial IGF1R signaling leads to decreased nitric oxide-dependent component of NVC responses, highlighting the importance of IGF-1 in maintaining normal cerebromicrovascular endothelial health.

Treatment with the BCL-2/BCL-xL inhibitor senolytic drug ABT263/Navitoclax improves functional hyperemia in aged mice

Stefano Tarantini et al.

Summary: The present study found that NVC responses were significantly impaired in aged mice, and treatment with ABT263/Navitoclax improved NVC response and hippocampal-encoded functions of learning and memory. The findings suggest that senescent cells in the aged brain may contribute to age-related neurovascular uncoupling, exacerbating cognitive decline. The neurovascular protective effects of ABT263/Navitoclax treatment highlight the potential of senolytic treatments as effective interventions in patients at risk for vascular cognitive impairment.

Summary: The study aims to summarize the evidence of CBF changes in normal aging and cognitive disorders. Age-related CBF changes may be related to progressive neuronal loss or diminished activity in the brain. Research suggests that CBF reduction might precede cognitive decline in VCI and AD.
