Characterization and in vitro testing of newly isolated lytic bacteriophages for the biocontrol of Pseudomonas aeruginosa

Liliam K. Harada et al.

Summary: In this study, two lytic phages that infect multidrug-resistant P. aeruginosa strains were isolated and characterized. Increasing the multiplicity of infection (MOI) significantly improved phage efficiency and delayed bacterial regrowth. Neither of the phages carried toxin, virulence, antibiotic resistance, or integrase genes. These findings are highly relevant in the context of bacterial resistance to antibiotics.

This study summarizes recent data on antibiotic resistance in priority HAIs (ESKAPE-E) in low- and lower-middle-income countries (L-LMICs) and compares them with data from high-income countries (HICs). The results show that antibiotic resistance is higher in L-LMICs compared to HICs.

Evaluation of Bacteriophage Cocktail on Septicemia Caused by Colistin-Resistant Klebsiella pneumoniae in Mice Model

Aprajita Singh et al.

Summary: This study aimed to optimize the dosage and frequency of bacteriophage cocktail for eradicating Klebsiella pneumoniae in septicemic mice. It was found that a lower dosage of 1 x 10(5) plaque-forming unit/mouse was effective in protecting the mice from fatal outcomes at any stage of septicemia. However, a higher dosage of 1 x 10(12) PFU/mouse was only fatal when given at the early hours of septicemia.

Summary: Fang et al. identified two previously unidentified phages that can inhibit the growth and decrease the virulence of carbapenem-resistant Klebsiella pneumoniae (CRKP). They also found that CRKP can develop phage resistance but can still be eliminated in a mouse intestinal colonization model, highlighting the potential of phage therapy as a treatment against drug-resistant pathogens.

Summary: The prolonged use of antibiotics has led to the development of multidrug resistance in many organisms, causing a global health crisis. Klebsiella pneumoniae is a causative organism for various infections and recently, both immunocompromised and healthy individuals are equally susceptible to K. pneumoniae infections due to the emergence of hypervirulent strains. This review discusses the pathophysiology of K. pneumoniae, including its virulence determinants, and explores potential drug targets for the discovery of novel drugs and effective treatment regimens.

Characterization of four virulent Klebsiella pneumoniae bacteriophages, and evaluation of their potential use in complex phage preparation

Summary: The studied bacteriophages in this work have a wide host range and high stability at different temperature and pH values. A cocktail of bacteriophages lysed all studied bacterial strains without the emergence of phage-resistant forms. Genomic data showed that the isolated viruses do not carry antibiotic resistance, virulence or lysogenic genes, and three out of four bacteriophages encode polysaccharide depolymerases.

Isolation and Characterization of Klebsiella Phages for Phage Therapy

Eleanor M. Townsend et al.

Summary: This study successfully isolated and characterized 30 novel, genetically diverse Klebsiella phages, belonging to six different phage families and nine genera. Among the Klebsiella-infecting phages, lytic phages were deemed most suitable for phage therapy due to their broad host range, high virulence, short lysis period, and lack of known toxin or antimicrobial resistance genes. It was found that a cocktail of multiple phages would be necessary to effectively treat Klebsiella infections due to the ease of Klebsiella to generate spontaneous phage-resistant mutants.
