Mining and engineering exporters for titer improvement of macrolide biopesticides in Streptomyces

Summary: Exporter engineering is a promising strategy for high-yield Streptomyces production in industrial biotechnology. A workflow for exporter mining was developed, along with a tunable plug-and-play exporter (TuPPE) module to optimize macrolide biopesticide production. By identifying three ATP-binding cassette transporters and utilizing a TuPPE module with replaceable promoters and ribosome binding sites, significant improvements in milbemycin titer were achieved. The use of the TuPPE module also led to enhanced titer levels in other Streptomyces strains, highlighting its potential for widespread applications in improving secondary metabolite production.

Summary: Propionic acid is commonly used in food and chemical industries, and its biotechnological production from biomass is seen as a more environmentally friendly alternative to traditional methods. In this study, key factors involved in PA tolerance in Pseudomonas putida KT2440 strain were identified through comparative transcriptional analysis. The MFS transporter gene cluster of PP_1271, PP_1272 and PP_1273 was found to play a crucial role in PA tolerance and its overexpression can improve tolerance in PA producing strains for potential use in engineering robust platforms for weak acid production.
