Advances allowing feasible pyrG gene editing by a CRISPR-Cas9 system for the edible mushroom Pleurotus eryngii

Tingli Wang et al.

Summary: A study identified a highly efficient gene editing system in the edible mushroom Pleurotus eryngii, which can generate various insertions and deletions through non-homologous end joining and homology-directed repair. By using a point-mutated variant of the endogenous sdhB gene as a selection marker, the system was successfully established for the first time, potentially opening up new possibilities for improving edible mushroom cultivars.

An Optimized and Efficient CRISPR/Cas9 System for the Endophytic Fungus Pestalotiopsis fici

Xinran Xu et al.

Summary: The study improved the genetic manipulation of endophytic fungus Pestalotiopsis fici by introducing the CRISPR/Cas9 system, achieving efficient genome editing such as site-specific gene insertion, dual-locus mutations, and long DNA fragment deletions. The method showed high efficiencies of up to 48.0% for site-specific gene insertion and 44.4% for two-site gene editing, while significantly shortening the genetic manipulation time for long DNA fragment deletion compared to traditional methods.

Hydrophobin CmHYD1 Is Involved in Conidiation, Infection and Primordium Formation, and Regulated by GATA Transcription Factor CmAreA in Edible Fungus, Cordyceps militaris

Xiao Li et al.

Summary: Hydrophobins are small proteins secreted exclusively by fungi, with Cmhyd1 being a member in Cordyceps militaris that plays various roles in fruiting body development, stress response, and regulation of transcription factors. Its deletion results in reduced aerial mycelia and conidiation, as well as inhibition of pigmentation and primordium differentiation. Cmhyd1 also participates in stress response and forms a positive feedback loop with Cmarea transcription factor. These findings provide insight into the diverse functions and regulatory mechanisms of Cmhyd1 in C. militaris.
