The study reveals the pivotal factors of immunosuppression in nccRCC tissues and their correlation with poor prognosis. Abnormal metabolic patterns were also found in both cancer cells and tumor-infiltrating stromal cells. The findings may contribute to the development of more effective therapies for nccRCC.

Single-cell transcriptomics reveals a low CD8+ T cell infiltrating state mediated by fibroblasts in recurrent renal cell carcinoma

Summary: This study investigated the differences in tumor microenvironment (TME) and gene expression patterns between primary renal cell carcinoma (pRCC) and recurrent renal cell carcinoma (reRCC) at the single-cell level. The results showed that reRCC had reduced CD8(+) T cells and increased infiltration of cancer-associated fibroblasts (CAFs), which were significantly associated with a worse response to immunotherapy. In addition, the study identified that CAFs expressed LGALS1 (Gal1), which could induce apoptosis of CD8(+) T cells and suppress tumor growth. Knockdown of Gal1 in CAFs increased CD8(+) T cell infiltration and enhanced the efficacy of immunotherapy.
