Nano delivery of simvastatin targets liver sinusoidal endothelial cells to remodel tumor microenvironment for hepatocellular carcinoma

Zhuo Yu et al.

Summary: This study found that the characteristics of liver sinusoidal endothelial cells (LSECs) are closely related to the development and prognosis of fibrotic HCC. Simvastatin restores the quiescence of activated hepatic stellate cells (aHSCs) by stimulating LSECs and upregulating the expression of CXCL16 in LSECs. LSEC-targeted delivery of simvastatin not only alleviates LSEC capillarization, but also inhibits tumor progression by recruiting natural killer T (NKT) cells through CXCL16. Conclusion: Simvastatin has an immune-based therapeutic effect on remodeling the immunosuppressive tumor microenvironment in HCC, providing a new strategy for HCC treatment.

Coagulation and Endothelial Dysfunction Associated with NAFLD: Current Status and Therapeutic Implications

Doris Ogresta et al.

Summary: Non-alcoholic fatty liver disease (NAFLD) is closely related to insulin resistance, type 2 diabetes mellitus, and obesity. It is considered a multisystem disease with a strong association with cardiovascular disease and arterial hypertension. Coagulation disorders are common in patients with hepatic impairment and are dependent on the degree of liver damage. NAFLD patients may have a procoagulant state, which may worsen liver injury.

Summary: Non-alcoholic steatohepatitis (NASH) is a serious transformation from simple steatosis to inflammation and hepatic injury. Profound metabolic dysregulations are associated with NASH, and the peroxisomal-proliferator-activated receptor alpha (PPAR alpha) plays a key role in its pathogenesis. However, many aspects of NASH have not yet been fully clarified.

The Scavenger Function of Liver Sinusoidal Endothelial Cells in Health and Disease

It also discusses the similarities and differences between LSECs and specialized scavenger endothelial cells (SECs) in various vascular beds and vertebrate classes. The activity of LSECs and other SECs helps to maintain blood and tissue homeostasis by efficiently clearing large molecules and nanoparticles, but this process may also result in unwanted uptake of nanomedicines and biologics.

Atorvastatin improves the sinusoidal contractile phenotype, thus enhancing endothelial function, while ambrisentan prevents HSC contraction by blocking ET-1 response. This combination treatment effectively decreases serum alanine aminotransferase levels and restores the quiescent phenotype of HSCs.
