Molecular characteristics and spatial distribution of adult human corneal cell subtypes

Summary: Bulk RNA sequencing captures the overall gene expression profile of a tissue, while single-cell RNA sequencing identifies distinct cell signatures based on transcriptomic identities. By analyzing single-cell RNAseq data from six adult human corneas, 16 cell clusters were bioinformatically identified, confirming the presence of stromal keratocytes, endothelium, various corneal epithelium subtypes, conjunctival epithelium, and supportive cells in the limbal stem cell niche. Further characterization revealed enriched biological pathways and molecular characteristics, providing novel insights into development, function, and location within the epithelial layer. The single-cell map of the human cornea enhances our understanding of its cellular subsets at a whole genome transcriptional level, with potential applications in normal corneal biology, disease pathology, and therapeutic approaches.
