Copper induces cell death by targeting lipoylated TCA cycle proteins

Peter Tsvetkov et al.

Summary: Copper is an essential cofactor for all organisms, but excessive copper can lead to cell death. We found that copper-dependent cell death mechanism is different from known mechanisms, and it is related to mitochondrial respiration. Copper directly binds to lipoylated components of the tricarboxylic acid cycle, causing protein aggregation and loss of iron-sulfur cluster proteins, ultimately leading to cell death.

Oxiapoptophagy: A type of cell death induced by some oxysterols

Thomas Nury et al.

Summary: Oxysterols, oxidized forms of cholesterol, induce cell death through oxidative stress, apoptosis, and autophagy at cytotoxic concentrations. This process involves activation of mitochondrial pathway, changes in mitochondrial permeability, and alterations in peroxisome function, ultimately leading to cell damage.

A decade of cell death studies: Breathing new life into necroptosis

Imran Khan et al.

Summary: Programmed cell death (PCD) has been a major research interest in the past three decades. Initially, apoptosis was considered the only mechanistic pathway for PCD, but now other important pathways such as necroptosis have been discovered, which are crucial for understanding the cellular processes behind different pathological conditions. Additionally, research on triggering necroptosis in cancer cells to induce cell death and overcome chemoresistance is being discussed.

Ca2+ overload- and ROS-associated mitochondrial dysfunction contributes to δ-tocotrienol-mediated paraptosis in melanoma cells

Michela Raimondi et al.

Summary: The study demonstrates that delta-tocotrienol induces paraptotic cell death in human melanoma cells, causing endoplasmic reticulum dilation and mitochondrial swelling. These alterations lead to impaired mitochondrial function, ROS production, and calcium overload.

Atg9-centered multi-omics integration reveals new autophagy regulators in Saccharomyces cerevisiae

Di Peng et al.

Summary: A multi-omic profiling revealed the influence of Atg9 on gene expression during autophagy and predicted 42 potential new Atg9-interacting proteins involved in autophagy. Two of these proteins, Glo3 and Scs7, were validated to be functional in autophagy, with Glo3 regulating the retrograde transport of Atg9. The study highlighted the importance of the Atg9 interactome and introduced a new method for analyzing multi-omics data.

Gasdermin D pore structure reveals preferential release of mature interleukin-1

Shiyu Xia et al.

Summary: Inflammasomes activate caspase-1 to cleave GSDMD and generate mature cytokines like IL-1 beta and IL-18, which are released through pores formed by GSDMD. The GSDMD pore mediates IL-1 release by electrostatic filtering, favoring the passage of mature cytokines over their precursors.

DHODH-mediated ferroptosis defence is a targetable vulnerability in cancer

Chao Mao et al.

Summary: The study reveals the mechanism of ferroptosis induced by inhibiting GPX4 in cancer cells, as well as the potential pathways to enhance or inhibit ferroptosis by intervening in relevant metabolic pathways. Therapeutic strategies targeting different levels of GPX4 expression could be a promising new approach for cancer treatment.

The lysosome as a master regulator of iron metabolism

Francesca Rizzollo et al.

Summary: This passage discusses the role of intracellular iron in essential cellular processes, the central role of lysosomes in maintaining iron homeostasis, and the regulatory circuits linking the lysosomal system to iron trafficking control, interorganellar crosstalk, and ferroptosis induction. It also explores how dysregulated lysosomal iron-handling functions contribute to diseases and can be utilized for therapeutic interventions.
