Cerebellar Contributions to Motor Impairments in People with Multiple Sclerosis

Alexandra C. Fietsam et al.

Summary: Recent studies have shown a clear association between cerebellar pathology and motor impairments in PwMS, but further research is needed to fully understand the pathophysiology. Future studies are necessary to determine the most effective treatments for motor impairments, including specific types and durations of exercise interventions and methods to enhance their effects.

Olig2 defines a subset of neural stem cells that produce specific olfactory bulb interneuron subtypes in the subventricular zone of adult mice

Oligodendrocytes and myelin: Active players in neurodegenerative brains?

Jing-Fei Chen et al.

Summary: Oligodendrocytes and myelin undergo dynamic changes in healthy and neurodegenerative brains, and may play an important role in modulating neuronal function and survival. Targeting oligodendroglial cells could have potential value in treating neurodegenerative diseases.

New oligodendrocytes exhibit more abundant and accurate myelin regeneration than those that survive demyelination

Sarah A. Neely et al.

Summary: Through research on zebrafish, it was found that surviving oligodendrocytes are less efficient in generating new myelin sheaths and frequently misdirect the myelin to neuronal cell bodies, which is similar to the pathology seen in multiple sclerosis (MS). Conversely, oligodendrocytes generated after demyelination produce abundant and correctly targeted myelin sheaths, suggesting that they may also have a better regenerative potential in MS.

Estimation of the density of neural, glial, and endothelial lineage cells in the adult mouse dentate gyrus

Joshua D. Rieskamp et al.

Summary: The study estimated the cell density of neural stem/progenitor cells in the dentate gyrus of adult mice using immunohistochemical methods and found that they had similar density as other cell types with secretory functions. These findings contribute to refining hypotheses about the roles of these cell types in regulating hippocampal function and their potential therapeutic uses.

Comparison of the Effects of Cuprizone on Demyelination in the Corpus Callosum and Hippocampal Progenitors in Young Adult and Aged Mice

Kyu Ri Hahn et al.

Summary: Feeding mice a diet enriched with cuprizone can induce demyelination in the corpus callosum and increase neural progenitor cells in the dentate gyrus. This damage is more pronounced in young adult mice and may be associated with changes in BDNF and pCREB levels.

GLI3 Is Required for OLIG2+Progeny Production in Adult Dorsal Neural Stem Cells

Rebecca J. Embalabala et al.

Summary: The ventricular-subventricular zone (V-SVZ) is a postnatal germinal niche that holds neural stem cells (NSCs) generating neurons and oligodendrocytes. Sonic hedgehog (Shh) signaling is essential for neurogenesis. This study investigates the role of transcription factors GLI2 and GLI3 in the proliferation and cell fate of dorsal and ventral V-SVZ NSCs.

Regional Cerebellar Volume Loss Predicts Future Disability in Multiple Sclerosis Patients

Katrin Parmar et al.

Summary: Through longitudinal imaging data of 163 MS patients, it was found that cerebellar volumes are associated with clinical scores, particularly motor and cognitive impairments. Regional cerebellar volume reduction is correlated with motor and cognitive disability in MS, serving as a predictor for future disease progression.

Neural stem cell pools in the vertebrate adult brain: Homeostasis from cell-autonomous decisions or community rules?

Nicolas Dray et al.

Summary: Adult stem cell populations must coordinate their own maintenance and the generation of differentiated cell types in a spatially controlled manner to sustain organ physiology. Feedback mechanisms coordinate stem cell maintenance and progeny generation in epithelia, while little is known about potential community processes underlying neural stem cell maintenance. Recent work on neural stem cell pools suggests that homeostasis may rely on population asymmetry and long-term spatiotemporal coordination of neural stem cell states and fates, indicating neural stem cell assemblies in the vertebrate brain behave as self-organized systems.

Reconditioning the Neurogenic Niche of Adult Non-human Primates by Antisense Oligonucleotide-Mediated Attenuation of TGFβ Signaling

Sebastian Peters et al.

Summary: Adult neurogenesis is a potential target for brain rejuvenation and regeneration. The use of a novel LNA-antisense oligonucleotide, NVP-13, has been shown to downregulate TGF beta-RII and enhance neurogenesis in both in vitro human neuronal precursor cells and in vivo non-human primates. This new approach holds promise for treating neurodegenerative disorders by modulating neurogenesis through targeting TGF beta pathways.

Regulation of Adult Mammalian Neural Stem Cells and Neurogenesis by Cell Extrinsic and Intrinsic Factors

Shuzo Matsubara et al.

Summary: Tissue-specific stem cells generate new functional cells to maintain tissue homeostasis and repair damaged tissue. Neurogenesis of endogenous NSCs alone is insufficient for complete brain repair, leading to exploration of direct reprogramming as an innovative approach. This review discusses the control of NSCs and neurogenesis in the adult brain, as well as the potential of direct neuronal reprogramming for future clinical applications.

Single-cell analysis of the ventricular-subventricular zone reveals signatures of dorsal and ventral adult neurogenesis

Arantxa Cebrian-Silla et al.

Summary: By analyzing a single-cell RNA-sequencing dataset of the adult mouse V-SVZ, two distinct populations of NSPCs residing in either the dorsal or ventral V-SVZ were identified. These regional differences in gene expression were validated using a single-nucleus RNA-sequencing reference dataset and immunocytochemistry. The study provides novel markers and territories for understanding the region-specific regulation of adult neurogenesis.
