Hif-1a suppresses ROS-induced proliferation of cardiac fibroblasts following myocardial infarction

Vaibhao Janbandhu et al.

Summary: Cardiac fibroblasts (CFs) and mesenchymal progenitors are more hypoxic and express more HIF-1a, leading to increased glycolytic metabolism. CF-specific deletion of Hif-1a results in increased mesenchymal progenitors and CF activation in uninjured hearts, but increased CF proliferation and excessive scarring after myocardial infarction. CF proliferation is associated with higher reactive oxygen species (ROS).

Pseudohypoxic HIF pathway activation dysregulates collagen structure-function in human lung fibrosis

Christopher J. Brereton et al.

Summary: This study provides evidence that the activation of the hypoxia-inducible factor (HIF) pathway is a critical regulator of pathogenetic collagen structure-function in fibrosis. Oxidative stress can drive the activation of pseudohypoxic HIF pathway, which alters collagen nanostructure and increases tissue stiffness.

TGF-β1 induced autophagy in cancer associated fibroblasts during hypoxia contributes EMT and glycolysis via MCT4 upregulation

Bikash Chandra Jena et al.

Summary: This study reveals the important role of Transforming growth factor-beta 1 (TGF-beta 1) in the tumor microenvironment and its interaction with cancer-associated fibroblasts (CAFs). The research shows that hypoxia and TGF-beta 1 induce autophagy in CAFs, leading to the supply of lactate to cancer cells and upregulation of aerobic glycolysis and monocarboxylate transporter (MCT4) expression in CAFs. Autophagy is also involved in the epithelial-mesenchymal transition (EMT) in hypoxic CAFs, promoting tumor migration and metastasis through the reverse Warburg effect.

DIVERSITY AND BIOLOGY OF CANCER-ASSOCIATED FIBROBLASTS

Summary: Efforts to develop anti-cancer therapies have mainly targeted the epithelial compartment, but recent studies have shown the significant influence of cancer-associated fibroblasts (CAFs) in tumor progression. CAFs not only promote cancer cell proliferation, therapy resistance, and immune exclusion, but may also restrain tumor progression in certain contexts. Research on CAFs has focused on their heterogeneity, plasticity, and functions across different cancer types and states, as well as advancements in therapeutic strategies targeting CAFs currently undergoing preclinical and clinical evaluation.

Exosomal circEIF3K from cancer-associated fibroblast promotes colorectal cancer (CRC) progression via miR-214/PD-L1 axis

Kaihua Yang et al.

Summary: In this study, a novel axis circEIF3K/miR-214/PD-L1 was revealed to mediate hypoxia-induced CRC progression via CAF, providing a rationale for developing new targeted therapeutics to treat CRC. The study found that low expression of circEIF3K was associated with prolonged survival time in normal tissues, and PD-L1 was confirmed as an important target for miR-214 in CRC.

Fibroblasts: Origins, definitions, and functions in health and disease

Maksim Plikus et al.

Summary: Fibroblasts are diverse mesenchymal cells that play a role in tissue homeostasis and disease, producing complex extracellular matrix and creating signaling niches. They are transcriptionally and functionally heterogeneous across and within organs, encoding positional information and maintaining distinct cellular progeny. Their unique properties make them poised for tissue repair but can also drive fibrotic disorders if activated aberrantly.

Perspectives on Hypoxia Signaling in Tumor Stroma

Yuqing Zhang et al.

Summary: Hypoxia is a common feature of solid tumors that promotes tumor progression and metastasis. Hypoxia, mainly mediated by the HIF family, leads to tumor cell migration, proliferation, metabolic changes, and therapy resistance. Additionally, hypoxia affects various cellular and non-cellular compartments in the tumor stroma, including disordered tumor vasculature and ECM homeostasis. Hypoxia also has complex and sometimes contradictory effects on immune cell function, contributing to an immunosuppressive environment.

Hypoxia-stimulated ATM activation regulates autophagy-associated exosome release from cancer-associated fibroblasts to promote cancer cell invasion

Lei Xi et al.

Summary: Studies have shown that oxidized ATM phosphorylates BNIP3 to induce autophagosome accumulation and exosome release in breast CAFs. In addition, oxidized ATM phosphorylates ATP6V1G1, leading to lysosomal dysfunction and facilitating exosome release through fusion with multi-vesicular bodies. This process ultimately stimulates non-canonical NF-kappa B signaling and enhances the invasive abilities of recipient breast cancer cells.

Hypoxia increases melanoma-associated fibroblasts immunosuppressive potential and inhibitory effect on T cell-mediated cytotoxicity

Linda Ziani et al.

Summary: This study reveals that hypoxia increases the expression and secretion of immunosuppressive factors by melanoma-associated fibroblasts, with hypoxic CAF secretome exerting a more profound effect on T cell-mediated cytotoxicity than normoxic counterpart. The crosstalk between hypoxia and CAFs may be an important determinant in the complex immunosuppressive tumor microenvironment.
