Augmented secretion of IL-1α from mouse oral squamous cell carcinoma (OSCC) vcells caused by serum deprivation and hypoxia promotes immune suppressive activity of mesenchymal stromal cells

Akihiro Matsunami et al.

Summary: The study found that IL-1 alpha production by Sq-1979-1 cells was synergistically enhanced under low serum and hypoxic conditions. The IFN-gamma-producing capability of stimulated spleen cells co-cultured with 10T1/2 cells was significantly reduced in the conditioned medium from Sq-1979-1 cells prepared with lower serum or under hypoxia. The expression of IL-1 alpha in OSCC tissues was prominent in the midst of a carcinomatous cellular lesion or a nearby necrotic lesion.

Association Between Interleukin-6 and Head and Neck Squamous Cell Carcinoma: A Systematic Review

Uzdan Uz et al.

Summary: This systematic review evaluates the activity of IL-6 in patients with head and neck squamous cell carcinoma (HNSCC). The findings suggest that elevated IL-6 concentrations in patients with HNSCC are associated with poor survival and oncological outcomes. Current IL-6 targeted therapies are mainly conducted in vitro and experimental studies, highlighting the need for prospective, randomized clinical trials to assess their effectiveness in treating HNSCC.

Summary: The study found that increased levels of IL-22 and IL-22R1 in laryngeal squamous cell carcinoma may be associated with the pathogenesis and prognosis of the cancer. This suggests that IL-22 could be an important biomarker that requires further research.

Role of interleukin-17 in human papillomavirus infection and associated malignancies

Utkarsha Sahu et al.

Summary: Human papillomavirus infection is a leading viral infection worldwide, primarily affecting the female genital tract and involving the host immune response. IL-17, secreted by Th17 cells, plays a significant role in enhancing disease severity in HPV infection and promoting cancer progression. Further research on the relationship between IL-17 and HPV infection pathology may be beneficial in developing therapeutic approaches.

Characterization of the immune response in patients with cancer of the oral cavity after neoadjuvant immunotherapy with the IRX-2 regimen

Siyu Liu et al.

Summary: IRX-2 is a homologous cell-derived multi-cytokine biologic with immune modulatory effects in oral cavity carcinoma patients, inducing changes in immune gene expression and DNA methylation. Immune-related genes showed alterations after IRX-2 therapy, with DMBT1 identified as a potential tumor suppressor. Overall, the effects of IRX-2 seem to have a subtle, targeted, or patient-specific impact.

Crosstalk between oral squamous cell carcinoma cells and cancer-associated fibroblasts via the TGF-β/SOX9 axis in cancer progression

Kenta Haga et al.

Summary: It was found that CAFs promote cancer cell migration and invasion through the TGF-beta/SOX9 axis, with SOX9 aiding in TGF-beta1-mediated invasion. Regional recurrence is significantly correlated with the presence of CAFs and SOX9 expression in invasive cancer nests.

AKT3 Is a Novel Regulator of Cancer-Associated Fibroblasts in Head and Neck Squamous Cell Carcinoma

Hideyuki Takahashi et al.

Summary: Cancer-associated fibroblasts (CAFs) play vital roles in tumor progression by promoting epithelial-to-mesenchymal transition, angiogenesis, and immunosuppression. This study identified AKT3, an AKT isoform, as a key regulator in CAFs in head and neck squamous cell carcinoma (HNSCC) with implications for tumor infiltration and prognosis. Targeting AKT3 in CAFs could potentially inhibit their pro-tumoral function and reverse CAF-mediated immunosuppression in cancer therapy.

The Role of Plasmacytoid Dendritic Cells in Cancers

Binhui Zhou et al.

Summary: Plasmacytoid dendritic cells (pDCs) are a special subtype of dendritic cells that produce massive amounts of type I interferon (IFN-I) and play a pivotal role in antiviral immunity. However, in many types of cancer, tumor-infiltrating pDCs do not respond to Toll-like receptor (TLR) stimulation, leading to reduced IFN-alpha production and promoting tumor growth.

The combined detection of Amphiregulin, Cyclin A1 and DDX20/Gemin3 expression predicts aggressive forms of oral squamous cell carcinoma

Ekaterina Bourova-Flin et al.

Summary: By analyzing the gene expression data of oral squamous cell carcinomas (OSCC), a combination of three genes associated with high-risk OSCC was identified, leading to the development of a prognostic stratification test. The study highlights the enhanced proliferative and poorly differentiated intrinsic nature of aggressive OSCC tumors. Experimental targeting of CCNA1 in OSCC cells resulted in a shift of transcriptomic signature towards a less aggressive form of OSCC, suggesting CCNA1 as a potential target for therapeutic approaches.

Improving function of cytotoxic T-lymphocytes by transforming growth factor-β inhibitor in oral squamous cell carcinoma

Yutaro Kondo et al.

Summary: Immunotherapy with immune-checkpoint therapy for OSCCs is limited by the immunosuppressive effects of TGF-beta, which inhibits CTL function; TGF-beta inhibitor effectively restores CTL function and is primarily expressed at the tumor invasive front; TGFB1 mRNA expression is significantly negatively correlated with the CD8(+) T-cell/Treg ratio and Ki-67 expression in CD8(+) T-cells, indicating TGF-beta suppresses CTL function in situ.

CXCL3 overexpression affects the malignant behavior of oral squamous cell carcinoma cells via the MAPK signaling pathway

Jinru Weng et al.

Summary: The study revealed that CXCL3 expression is higher in OSCC tumors compared to normal tissues, and is closely associated with tumor stage and lymph node metastasis. Experiments showed that exogenous recombinant human CXCL3 and HSC-4 cells overexpressing CXCL3 enhanced proliferation and migration abilities. In vivo xenograft experiments demonstrated that CXCL3 overexpression promotes tumor growth. Further analysis revealed that the ERK1/2 blocker PD98059 can attenuate the proliferation and migration effects induced by CXCL3.
