The secretome of stressed peripheral blood mononuclear cells increases tissue survival in a rodent epigastric flap model

Stefan Hacker et al.

Summary: The study investigated the beneficial effects of a secretome derived from gamma-irradiated PBMCs on tissue survival, angiogenesis, and clinical parameters after flap surgery in a rodent epigastric flap model. Significant reductions in tissue necrosis rate and increased vessel counts were observed with PBMCsec treatment, suggesting its potential as a therapeutic approach in reconstructive surgery. Seroma formation was also decreased after treatment with fibrin sealant with or without the addition of PBMCsec, indicating improved outcomes post-surgery.

Basophil activation test: Mechanisms and considerations for use in clinical trials and clinical practice

Alexandra F. Santos et al.

Summary: The basophil activation test (BAT) measures the degree of degranulation of basophils following stimulation with allergen, and it can be used as a more specific diagnostic tool for allergic diseases. Clinical application of BAT requires clinical validation, quality assurance, and efforts are ongoing to establish a platform for laboratories to ensure reproducibility and reliability of results.

Safety and clinical efficacy of the secretome of stressed peripheral blood mononuclear cells in patients with diabetic foot ulcer-study protocol of the randomized, placebo-controlled, double-blind, multicenter, international phase II clinical trial MARSYAS II

Alfred Gugerell et al.

Summary: Diabetes and its complications pose a growing health risk worldwide, leading to a desperate need for effective and safe treatments. The study aims to evaluate the efficacy of the novel drug APOSEC in promoting healing of diabetic foot ulcers through a randomized, double-blind, placebo-controlled trial involving 132 patients.

Comparing the efficacy of γ- and electron-irradiation of PBMCs to promote secretion of paracrine, regenerative factors

Maria Laggner et al.

Summary: The use of electron-irradiation as an alternative radiation source for producing therapeutic cell-free secretomes has been demonstrated to be effective, with comparable effects on PBMCs and similar regenerative potential to gamma-irradiation. The secretion patterns and biological activities of secretomes generated from both radiation sources showed high similarity, suggesting the potential for future clinical trials in tissue-regenerative medicine.
