Molecular Mechanisms Underlying Neurotransmitter Release

Josep Rizo

Summary: Recent advances in understanding the mechanism of neurotransmitter release have revealed a model involving SNARE proteins, NSF, SNAPs, Munc18-1, Munc13-1, synaptotagmin-1, and complexin. These proteins interact in a tightly regulated pathway to ensure membrane fusion and release of neurotransmitters, providing insights into presynaptic plasticity mechanisms.

Control of neurotransmitter release by two distinct membrane-binding faces of the Munc13-1 C1C2B region

Marcial Camacho et al.

Summary: The C-1-C2B region of Munc13-1 plays a central role in neurotransmitter release, with distinct faces controlling release and short-term presynaptic plasticity. Mutations in the polybasic face severely impair Ca2+-independent liposome bridging and fusion, while mutations in Ca2+-binding loops have milder effects and can enhance or impair Ca2+-evoked release.

Phosphatidylinositol and phosphatidylinositol-3-phosphate activate HOPS to catalyze SNARE assembly, allowing small headgroup lipids to support the terminal steps of membrane fusion

Thomas Torng et al.

Summary: Intracellular membrane fusion requires a combination of specific lipids such as phosphatidylinositol and phosphatidylinositol-3-phosphate for HOPS activation and other lipids like phosphatidylethanolamine, phosphatidic acid, phosphatidylserine, and diacylglycerol for the terminal bilayer rearrangement step of fusion. Different lipid requirements are crucial for activating HOPS for trans-SNARE assembly and for the final fusion process.
