Osteon: Structure, Turnover, and Regeneration

Bei Chang et al.

Summary: This review highlights the importance of osteons in bone mechanics and turnover, emphasizing the crucial role of osteon structure in cortical bone regeneration. It provides insights into the hierarchical structure and critical functions of osteons, as well as the modeling and remodeling processes at a cellular level. The review also introduces bioengineering approaches developed to mimic the osteon structure, aiming to attract attention in the orthopedic community for future cortical bone regeneration research.

Effect of geometrical structure variations on the viscoelastic and anisotropic behaviour of cortical bone using multi-scale finite element modelling

Ampaiphan Atthapreyangkul et al.

Summary: Multi-scale finite element analysis was conducted to investigate the impact of geometrical changes on the mechanical properties of cortical bone at different structural scales. The study incorporated statistical approaches and sensitivity analyses, revealing an exponential correlation between mineral volume fraction and effective stiffness constants at each length scale. Fast and slow response relaxation behavior of cortical bone was found to be influenced by bone's structural anisotropy and global mineral volume fraction variations between length scales.

Osteoclasts recycle via osteomorphs during RANKL-stimulated bone resorption

Michelle M. McDonald et al.

Summary: Research has shown that RANKL-stimulated osteoclasts can undergo fission to form osteomorphs, which are involved in the regulation of bone resorption and could be targeted for the treatment of skeletal diseases. Single-cell RNA sequencing revealed that osteomorphs are transcriptionally distinct from osteoclasts and macrophages, expressing non-canonical osteoclast genes associated with bone phenotypes when deleted in mice.

Single-Cell RNA-Sequencing Reveals the Breadth of Osteoblast Heterogeneity

Hirotaka Yoshioka et al.

Summary: Our study identified four clusters of osteoblasts based on gene expression, with three showing similar expression of osteoblast markers and one being distinct. The cells in these clusters exhibited heterogeneity in gene expression, independent of maturational stage, suggesting diversity in functional activities and developmental fates. The analysis also revealed genes involved in protein synthesis and trafficking between endoplasmic reticulum and Golgi as active in these clusters.

Current and emerging histomorphometric and imaging techniques for assessing age-at-death and cortical bone quality

Janna M. Andronowski et al.

Summary: Bones are dynamic living organs that undergo continual change throughout life. High-resolution imaging techniques have the potential to provide new information about changes in bone microarchitecture. Methodological approaches used historically by researchers to assess the products of remodeling are continuously evolving.
