PHYSIOLOGY AND PATHOPHYSIOLOGY OF HUMAN AIRWAY MUCUS

David B. Hill et al.

Summary: The mucus clearance system is the dominant mechanical defense system of the human lung, clearing mucus through cilia and airflow. Mucus transport rates are heavily dependent on mucus concentration and can be accurately predicted by a model. Fluid for mucus hydration is generated through epithelial fluid transport. Diseases can cause mucus hyperconcentration and obstructive plaques.

Sterol regulation of developmental and oncogenic Hedgehog signaling

Vikas Daggubati et al.

Summary: This article discusses the role of the Hedgehog (Hh) family of lipid-modified signaling proteins in embryonic tissue patterning and postembryonic tissue homeostasis, as well as the involvement of dysregulated Hh signaling in familial and sporadic cancers. It also explores the binding and modulation of SMO activity by endogenous cholesterol and oxidized cholesterol derivatives. The article proposes models of sterol regulation of SMO and discusses the potential utility of steroidal SMO ligands or inhibitors of enzymes involved in sterol metabolism as cancer therapeutics.

Insulin-degrading enzyme ablation in mouse pancreatic alpha cells triggers cell proliferation, hyperplasia and glucagon secretion dysregulation

Beatriz Merino et al.

Summary: This study found that loss of IDE function in alpha cells leads to impaired alpha cell function, resulting in abnormal glucagon secretion and hyperglucagonaemia. The study also revealed that IDE deficiency in alpha cells impairs the ability to suppress glucagon release in the presence of high glucose or insulin, and contributes to alpha-synuclein aggregation which may affect glucagon secretion via cytoskeletal dysfunction.

LPA signaling acts as a cell-extrinsic mechanism to initiate cilia disassembly and promote neurogenesis

Huai-Bin Hu et al.

Summary: Dynamic assembly and disassembly of primary cilia play critical roles in tissue development and homeostasis. The authors have identified lysophosphatidic acid (LPA) as a physiological extracellular factor that initiates cilia disassembly through both YAP/TAZ mediated transcription and calcium/calmodulin mediated activation of Aurora A.

Additionally, the study suggests the existence of a feedback loop between primary cilia and the canonical Wnt pathway.

The structural basis of intraflagellar transport at a glance

Mareike A. Jordan et al.

Summary: The intraflagellar transport (IFT) system is a crucial molecular machine used by cells to assemble and maintain the cilium, shuttling structural components and signaling receptors within the organelle. IFT trains, composed of IFT-A and IFT-B adaptor protein complexes, are powered by kinesin-2 and IFT-dynein motors for bidirectional movement along microtubules.

Co-Adaptation of Physical Attributes of the Mammalian Female Reproductive Tract and Sperm to Facilitate Fertilization

Chih-Kuan Tung et al.

Summary: The female reproductive tract plays multiple essential roles in the reproductive process, supporting the development and transport of fertilized eggs while protecting against invasive pathogens. Sperm are considered pusher microswimmers, propelled by pushing fluid, and their migration is influenced by the architecture and flow characteristics of the tract. The viscoelasticity of fluids in the tract improves sperm cooperation and prevents pathogen invasion.
