Desilylation Induced by Metal Fluoride Nanocrystals Enables Cleavage Chemistry In Vivo

Dongban Duan et al.

Summary: Metal fluoride nanocrystals can induce controlled release of fluorophores and drug molecules in various biological systems through desilylation cleavage chemistry. The use of biocompatible PEG-coated CaF2 nanocrystals has shown efficient release of functional molecules. This strategy has also been applied to prodrug activation with remarkable anticancer effects and minimal side effects, empowering metal fluoride nanocrystals for further biological applications.

Rapid Two-Photon Fluorescence Imaging of Monoamine Oxidase B for Diagnosis of Early-Stage Liver Fibrosis in Mice

Nannan Fan et al.

Summary: This study established a two-photon fluorescence imaging method for in vivo detection of MAO-B activity using a simply prepared probe, BiPhAA. The platform is more rapid than other current detection methods, allowing dynamic observation of endogenous MAO-B level changes in hepatic stellate cells. The BiPhAA-based imaging platform successfully distinguished mice with liver fibrosis from normal mice by showing six times higher fluorescence brightness in the liver tissue of fibrosis mice.

Dual-locked spectroscopic probes for sensing and therapy

Luling Wu et al.

Summary: Optical imaging probes provide a non-invasive and longitudinal way to detect and uncover the physiological and pathological functions of an analyte of interest at the molecular level. Dual-locked systems offer enhanced specificity and multiplex detection, allowing for crosstalk-free fluorescent and chemiluminescent detection of two distinct biomarkers, making them particularly interesting in the field of research.

A trifunctional self-immolative spacer enables drug release with two non-sequential enzymatic cleavages

Alberto Dal Corso et al.

Summary: The cyclative cleavage of an amine-carbamate self-immolative spacer for releasing a hydroxyl cargo was hindered by the introduction of a phosphate monoester handle in the spacer. This trifunctional spacer was incorporated into a model anticancer prodrug, which exhibited rapid drug release only in the presence of both a protease and a phosphatase enzyme.
