Mannose enhances the radio-sensitivity of esophageal squamous cell carcinoma with low MPI expression by suppressing glycolysis

Hui Luo et al.

Summary: This study investigated the effect of mannose on the radio-sensitivity of human esophageal squamous cell carcinoma (ESCC) cell line and its possible mechanism. The results showed that mannose had inhibitory effects on both KYSE70 and KYSE450 cell lines. The inhibitory effect was more significant on the KYSE450 cell line with an increased mannose concentration. Furthermore, mannose could enhance the radio-sensitivity and apoptosis rate of KYSE450 cell line, while its efficacy on KYSE70 cell line was limited. Intracellular metabolite analysis revealed that mannose could disturb glycolysis when combined with radiation therapy in esophageal cancer cells.

Summary: Every year, the American Cancer Society projects the numbers of new cancer cases and deaths in the United States, with the latest data showing a significant decline in lung cancer mortality, while prostate cancer mortality has plateaued and breast and colorectal cancer mortality have slowed. Improvements in treatment have accelerated progress against lung cancer, leading to a record drop in overall cancer mortality.

Ferroptosis: mechanisms, biology and role in disease

Xuejun Jiang et al.

Summary: Ferroptosis, as a form of regulated cell death driven by iron-dependent phospholipid peroxidation, has seen significant growth in research in recent years. Studies have focused on molecular mechanisms, regulation, and functions of ferroptosis, linking this cell death modality to various pathologies and proposing its roles in normal physiology and potential therapeutic targeting.

Ferroptosis becomes immunogenic: implications for anticancer treatments

Daolin Tang et al.

Summary: Ferroptosis is an iron-dependent form of non-apoptotic cell death that has been shown to have antitumor immune effects, inducing immunogenic cell death by emitting DAMPs and triggering dendritic cell maturation in vitro. Furthermore, ferroptotic cells are capable of vaccinating against fibrosarcoma rechallenge in preclinical models.

Broadening horizons: the role of ferroptosis in cancer

Xin Chen et al.

Summary: Ferroptosis is an iron-dependent form of regulated cell death driven by excessive lipid peroxidation, and can be induced by various agents to suppress tumor growth while also potentially triggering inflammation-associated immunosuppression. The extent of ferroptosis's impact on tumor biology and its interactions with other signaling pathways are still under investigation.

Mechanisms of stearoyl CoA desaturase inhibitor sensitivity and acquired resistance in cancer

Nicole Oatman et al.

Summary: In glioblastoma, some patients have low levels of SCD expression, leading to the evolution of cell lines that can survive independent of SCD, despite unknown mechanisms. Cell lines that escape genetic and epigenetic alterations become highly dependent on SCD for survival. Resistance to SCD inhibition in SCD-dependent lines is mediated through a previously unknown FOSB mechanism, which can be blunted to extend survival in tumor-bearing mice.

Immunogenic ferroptosis and where to find it?

Robin Demuynck et al.

Summary: Ferroptosis is a newly discovered form of regulated cell death distinct from apoptosis and necroptosis, and has potential in anticancer therapy. However, ferroptotic cancer cells may pose a risk of tumor evasion due to decreased phagocytosis and antigen cross-presentation. Factors such as lactate, acidification, and hypoxia in the tumor microenvironment play a role in promoting ferroptosis resistance and affecting its immunogenicity.

Ferroptosis as a mechanism to mediate p53 function in tumor radiosensitivity

Guang Lei et al.

Summary: The study reveals the critical role of ferroptosis in p53-mediated tumor radiosensitivity, promoting lipid peroxidation and cell death by inhibiting SLC7A11. P53 deficiency leads to radioresistance in cancer cells or tumors, while ferroptosis inducers that inhibit SLC7A11 significantly enhance tumor sensitivity to radiotherapy.

Holo-lactoferrin: the link between ferroptosis and radiotherapy in triple-negative breast cancer

Zheng Zhang et al.

Summary: The study showed that iron-saturated Lactoferrin (Holo-Lf) can induce ferroptosis in triple-negative breast cancer cells and enhance their sensitivity to radiotherapy. Conversely, low iron-saturated Lactoferrin (Apo-Lf) may inhibit this cell death process. Non-triple-negative breast cancer cells may have resistance to Holo-Lf-induced ferroptosis due to higher redox balance capacity.
