Ferritin reduction is essential for cerebral ischemia-induced hippocampal neuronal death through p53/SLC7A11-mediated ferroptosis

Summary: The study investigated the impact of lentivirus-induced ferritin overexpression on hippocampal neuronal injury and death in a rat model of cerebral ischemia-reperfusion. The results showed that ferritin overexpression partially reversed the cognitive and locomotor deficits induced by ischemic injury, reduced tau hyperphosphorylation, and protected against pathogenic changes, apoptosis, and oxidative stress in hippocampal neurons. The study also demonstrated that ferritin reduction is essential for preventing hippocampal neuronal ferroptosis mediated by p53 and SLC7A11 upregulation in response to cerebral ischemia.

Summary: This study demonstrated that arsenite can trigger ferroptosis in neuronal cells through activation of ferritinophagy. Regulating ferritinophagy to control iron levels may provide a clue for prevention against arsenite neurotoxicity. Co-treatment with ferrostatin-1 or autophagy inhibitors significantly attenuated the cytotoxic effects of arsenite.
