Reprogramming of H3K9bhb at regulatory elements is a key feature of fasting in the small intestine

Summary: The study shows that β-OHB is an important metabolic energy source during fasting and acts as a chromatin regulator through lysine β-hydroxybutyrylation. Enrichment of H3K9bhb at critical gene promoters associated with lipolytic and ketogenic metabolic pathways in the small intestine during fasting was observed. Fasting also results in an enrichment of an H3K9bhb-H3K27ac signature at active metabolic gene promoters and distal enhancer elements.

Summary: Knockout mice are widely used to study the phenotypic effects of gene dysfunction, and the use of RNA-guided Cas9 nuclease technology has significantly reduced the time and resources needed to produce knockout mouse lines. A standard methodology using electroporation of Cas9 ribonucleoprotein (RNP) into mouse zygotes has resulted in an 80% success rate in generating founders for null alleles with a subsequent 93% germline transmission rate. These methods rely on existing equipment in transgenic facilities and are relatively straightforward to implement with appropriate expertise in embryo handling.
