Microscale thermophoresis suggests a new model of regulation of cardiac myosin function via interaction with cardiac myosin-binding protein C

Saraswathi Ponnam et al.

Summary: The study reveals that cMyBP-C regulates the function of cardiac myosin by interacting with different regions of the myosin molecule. The N-terminal domains of cMyBP-C interact with the head and tail domains of myosin through phosphorylation-independent and phosphorylation-dependent interactions, while the highest affinity interaction sites between cMyBP-C and myosin S1 are located in the central domains.

The effects of L-arginine on protein stability and DNA binding ability of SaeR, a transcription factor in Staphylococcus aureus

Ruochen Fan et al.

Summary: The SaeRS two-component system in Staphylococcus aureus regulates the expression of virulence factors, and adding L-arginine to purification buffers can improve the stability and solubility of recombinant protein SaeR. The thermal denaturation temperature of SaeR is significantly increased in 10 mM L-Arg buffer, and MST analysis shows that SaeR can bind to the P1 promoter under different phosphorylation statuses in buffers containing L-Arg. These findings suggest an effective method for purifying SaeR and other proteins.

Strong interactions between Salp15 homologues from the tick I. ricinus and distinct types of the outer surface OspC protein from Borrelia

Paulina Bierwagen et al.

Summary: Ticks of the genus Ixodes are parasites that feed on vertebrate blood and act as vectors for various pathogenic microbes, including the causative agent of Lyme borreliosis. The saliva of these ticks contains bioactive molecules, with the Salp15 protein being a well-studied component that suppresses the host immune response and interacts with Borrelia burgdorferi spirochetes. Recent research has shown that in addition to Salp15, other homologues from Ixodes ricinus are also capable of binding to the outer surface protein C (OspC) of Borrelia burgdorferi, potentially aiding in evading the host immune system.
