Human IL-10-producing B cells have diverse states that are induced from multiple B cell subsets

Marla C. Glass et al.

Summary: Regulatory B cells (Bregs) secrete interleukin-10 (IL-10) to suppress immune responses, but there is no definitive immunophenotype to identify these B cells. Multiple functional subsets of B cells can produce IL-10, and there is no unique phenotype for IL-10(+) B cells. Furthermore, a significant portion of IL-10(+) B cells also express the pro-inflammatory cytokines IL-6 and tumor necrosis factor alpha (TNF-alpha). However, operationally tolerant liver transplant recipients have an enrichment of IL-10(+) but not TNF-alpha(+) or IL-6(+) B cells compared to those receiving immunosuppression. Therefore, IL-10-producing B cells in humans constitute an induced and transient state arising from various subsets of B cells, which may contribute to the maintenance of immune homeostasis.

Longitudinal analysis reveals high prevalence of Epstein-Barr virus associated with multiple sclerosis

Summary: The study found that in patients with multiple sclerosis, IL-10 produced by transitional B cells is significantly reduced, leading to the failure of suppressing effector T-cell function. This defect may contribute to the immune pathological process in multiple sclerosis.

Elevated Circulating IL-10 Producing Breg, but Not Regulatory B Cell Levels, Restrain Antibody-Mediated Rejection After Kidney Transplantation

Yongsheng Luo et al.

Summary: Circulating IL-10(+) Breg levels are a more appropriate indicator for assessing resistance to AMR after kidney transplantation compared to Bregs. Kidney B cell and IL-10 infiltration, as well as high expression of CXCL13, were associated with AMR. Furthermore, circulating IL-10(+) Bregs remained elevated during the 90-day post-operation period, contributing to immune homeostasis.

Role of B Cell Profile for Predicting Secondary Autoimmunity in Patients Treated With Alemtuzumab

Paulette Esperanza Walo-Delgado et al.

Summary: Baseline blood lymphocyte profile can help identify relapsing remitting multiple sclerosis (RRMS) patients at higher risk of developing secondary autoimmune adverse events (AIAEs) after alemtuzumab treatment. Patients with higher B-cell percentages and lower CD4+ T cell percentages at baseline were more likely to develop AIAEs.

Evolution of Human Memory B Cells From Childhood to Old Age

Michela Ciocca et al.

Summary: The decline of the immune system in aging leads to decreased susceptibility to infections and reduced response to vaccination, especially in reacting to new pathogens. Research shows that elderly individuals have a significant reduction in CD27(dull) memory B cells, while having a higher population of specialized antigen-selected CD27(bright) memory B cells. Additionally, B cells from older individuals produce significantly fewer IgM and IgA antibodies compared to younger individuals after in vitro stimulation.
