Integrative analysis from multi-center studies identities a consensus machine learning-derived lncRNA signature for stage II/III colorectal cancer

Summary: This study comprehensively identified recurrence-related lncRNAs and determined an optimal machine learning-derived lncRNA signature for predicting the risk of recurrence in colorectal cancer patients. The signature was independent of common clinical and molecular features and showed better performance than these variables. Further validation confirmed these findings and demonstrated that patients with a high signature benefited more from a certain chemotherapy drug, while patients with a low signature were more sensitive to another drug.

Summary: The upregulation of SOX2 plays a central role in the acquisition of the CAF phenotype, which is repressed by PKC zeta. The PKC zeta-SOX2 axis is crucial in controlling the pro-tumorigenic potential of CAFs, as loss of SOX2 or Sfrp1/2 in CAFs impairs the migration, invasion, and tumorigenicity of colon cancer cells. High stromal levels of PKC zeta and SOX2 expression in the stroma are associated with worse prognoses in colorectal cancer patients.

Additionally, a population of antigen-presenting macrophages interacting with CD8 T cells consistently segregated with response to anti-PD1 agents.

These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
