Single-cell multiomics reveals persistence of HIV-1 in expanded cytotoxic T cell clones

Summary: Understanding the driving factors and markers of clonally expanding HIV-1-infected CD4(+) T cells is crucial for the eradication of HIV-1. Utilizing single-cell ECCITE-seq technology, researchers tracked the dynamics of clonal expansion in longitudinally archived samples from HIV-1-infected individuals and uninfected individuals. The study revealed that persistent antigen and TNF responses shaped T cell clonal expansion, even in the presence of antiretroviral therapy. Additionally, HIV-1-infected cytotoxic CD4(+) T cell clones persisted and were enriched in cytotoxic effector memory Th1 cells.

Summary: Using single-cell transcriptomic analysis, researchers identified multiple subsets of CD4(+) T cells in the intestines of colitic mice, including a subset that produces interferon (IFN)-gamma. In vivo experiments showed that challenge with the food colorant Red 40 activated CD4(+) T cells and caused intestinal epithelial cell apoptosis, which was attenuated by depletion of CD4(+) cells and blocking IFN-gamma. Experiments conducted ex vivo demonstrated that intestinal CD4(+) T cells from colitic mice directly promoted apoptosis of intestinal epithelial cells and enteroids.

Tumor-specific cytolytic CD4 T cells mediate immunity against human cancer

Summary: CD4 T cells displaying cytotoxic phenotypes were identified in different human cancers through mining single-cell RNA-seq datasets. Ex vivo confirmation of the cytolytic tumor-specific CD4 T cells was achieved using peptide-MHCII-multimer technology. The cytotoxic activity of these cells, partially dependent on SLAMF7, was demonstrated to have delayed kinetics compared to classical cytotoxic lymphocytes, and agonistic engagement of SLAMF7 enhanced their cytotoxicity, indicating potential synergy with other cancer immunotherapies.

Cytotoxic CD4+T-cells specific for EBV capsid antigen BORF1 are maintained in long-term latently infected healthy donors

Summary: Epstein Barr Virus (EBV) infects a large percentage of the population and establishes latent infection in B-cells. Efforts to develop EBV prophylactic vaccination have mainly focused on neutralising antibodies, but generating T-cell responses against EBV structural proteins could be a promising alternative strategy. Research has shown that EBV capsid proteins BcLF1, BDLF1, and BORF1 are frequent targets of T-cell responses, and CD4+ T-cells specific for these proteins can recognize and control newly EBV-infected cells, even in long-term viral carriage. This study highlights the potential of targeting structural antigens for prophylactic EBV vaccine development.

Summary: Research found that CD4 CTL are resistant to suppression by Tregs in vitro, but the conditioned medium of CD4 CTL can accentuate the suppressive phenotype of Tregs. The CD4 CTL conditioned medium also skews memory TH cells towards a TH17 phenotype, and when cell-cell contact is established between CD4 CTL and TH cells, the proliferation of TH cells is no longer increased and Treg-mediated suppression is restored.

Higher densities of T cell subsets, B cells, and macrophages were observed in the central region of pMMR CRC tumor tissues. CD4(+) GzmB(+) T cells in the central region were found to be positively correlated with patient prognosis, and their infiltration was enhanced by neoadjuvant chemotherapy, improving overall survival and disease-free survival in pMMR CRC patients. Further research on the use of CD4(+) GzmB(+) T cells in tumor immunotherapy is warranted.
