Improvement of the anticancer efficacy of PD-1/PD-L1 blockade via combination therapy and PD-L1 regulation

Mengling Wu et al.

Summary: PD-1/PD-L1 immune checkpoints inhibit the anticancer effect of T cells in the tumor microenvironment, and downregulation or upregulation of PD-L1 expression can improve the efficacy of anti-PD-1/PD-L1 treatment. Overcoming the current limitations of this treatment can expand its anticancer applications and improve patient response rates and survival time.

The Heterogeneity of Immune Cell Infiltration Landscape and Its Immunotherapeutic Implications in Hepatocellular Carcinoma

Yuanyuan Guo et al.

Summary: Immunotherapy has a low response rate in hepatocellular carcinoma (HCC), but a study revealed that immune infiltration patterns and CTNNB1 mutation status can impact the efficacy of immunotherapy and prognosis in HCC patients. Evaluating immune infiltration patterns can improve understanding of the tumor immune microenvironment and provide new directions for individualized immunotherapy strategies for HCC.

An immune-related gene signature for predicting survival and immunotherapy efficacy in hepatocellular carcinoma

Yifei Dai et al.

Summary: A novel immune-related gene signature was developed to accurately predict survival and immunotherapy efficacy for HCC patients. Analysis of HCC samples identified 11 differentially expressed immune-related genes, which were used to establish the IRGPI with high predictive accuracy and ability to reflect the immune microenvironment and predict immunotherapy efficacy.

The Current Landscape of Immune Checkpoint Blockade in Hepatocellular Carcinoma: A Review

Matthias Pinter et al.

Summary: Recent advancements in the treatment of advanced hepatocellular carcinoma (HCC) have led to the approval of new systemic therapies such as angiogenesis inhibitors and immune checkpoint blockers, indicating important progress in the field. Strategies to target immunosuppression in tumors, such as vascular endothelial growth factor pathway inhibition and transforming growth factor-beta pathway inhibition, show promise in enhancing the efficacy of immunotherapy in HCC. Successes and failures of these treatments have provided insights into implementing novel systemic therapy in HCC and improving immunotherapy efficacy for this disease.

Adh4, an alcohol dehydrogenase controls alcohol formation within bacterial microcompartments in the acetogenic bacterium Acetobacterium woodii

Nilanjan Pal Chowdhury et al.

Summary: Acetobacterium woodii utilizes the Wood-Ljungdahl pathway for acetate production from CO2, but can also metabolize 1,2-propanediol to produce propionate and propanol. Adh4 in A. woodii is the alcohol dehydrogenase responsible for propanol formation within bacterial microcompartments, playing a crucial role in NADH recycling and redox balancing. Deletion of adh4 gene in A. woodii results in reduced propanol formation and altered metabolic end products.

Significant down-regulation of growth hormone receptor expression revealed as a new unfavorable prognostic factor in hepatitis C virus-related hepatocellular carcinoma

Ching-Chih Lin et al.

Summary: The study identified a significant down-regulation of GHR expression in HCV-related HCC tissues compared to ANT liver tissues. Lower GHR expression was associated with older age, as well as worse clinicopathologic characteristics, suggesting that decreased GHR expression levels contribute to the development of HCC.

Advances in immunotherapy for hepatocellular carcinoma

Bruno Sangro et al.

Summary: Immunotherapeutic interventions have shown effectiveness in the treatment of hepatocellular carcinoma. Checkpoint inhibitors have demonstrated strong anti-tumor activity, but other immune strategies have not yet shown consistent clinical activity. Advancing treatment to earlier stages, discovering predictive biomarkers, and exploring effective combinatorial approaches are key challenges in HCC immunotherapy.

An Inflammatory Response-Related Gene Signature Can Impact the Immune Status and Predict the Prognosis of Hepatocellular Carcinoma

Zhuo Lin et al.

Summary: Hepatocellular carcinoma (HCC) is a highly heterogeneous disease, and the prognostic prediction is challenging due to its complexity. In this study, a novel prognostic signature consisting of eight inflammatory response-related genes was constructed using LASSO Cox regression analysis. These genes were found to be correlated with overall survival, immune status, cancer-related pathways, and drug sensitivity in HCC patients.
