Hyocholic acid species improve glucose homeostasis through a distinct TGR5 and FXR signaling mechanism

Summary: This study reveals that high levels of hyocholic acid (HCA) in pigs contribute to their resistance against type 2 diabetes, as depletion of bile acids (BA) in pigs leads to reduced glucagon-like peptide-1 (GLP-1) secretion and increased blood glucose levels. HCA administration in diabetic mouse models effectively improves GLP-1 secretion and glucose homeostasis by activating TGR5 and inhibiting FXR in enteroendocrine cells, a mechanism not seen in other BA species. Clinical findings also show a correlation between lower levels of HCA species and diabetes, indicating its importance in glycemic control.

Alteration of gut microbial profile in patients with diabetic nephropathy

Summary: Investigations have shown that 30-40% of patients with diabetes develop diabetic nephropathy (DN). The gut microbiome has become a lively field of research in diabetes mellitus and chronic kidney disease. This study analyzed the gut microbial profiles in DN patients and healthy controls, revealing differences in microbial profiles based on disease stages, gender, and BMI. The results demonstrated dysbiosis and a decrease in gut bacterial richness and diversity in DN patients compared to healthy controls. Additionally, certain genera identified in the study may serve as potential microbial biomarkers for DN.

GPR43 deficiency protects against podocyte insulin resistance in diabetic nephropathy through the restoration of AMPK alpha activity

Summary: The study showed that GPR43 expression was increased in kidney samples of DN patients, diabetic animal models, and high glucose-stimulated podocytes, with deletion of GPR43 alleviating albuminuria and renal injury in diabetic mice. Antibiotic treatment and fecal microbiota transplantation improved podocyte insulin sensitivity and attenuated glomerular injury in diabetic rats by downregulating GPR43 expression and reducing serum acetate levels.
