Summary: Allergic immunity is regulated by ILC2s and Th2 cells at various barriers, and their dynamic repositioning during immune perturbations is observed. Allergic inflammation drives the invasion of lung and liver non-adventitial parenchyma by ILC2s and Th2 cells. Concurrent type 1 and type 2 mixed inflammation can block the parenchymal trafficking of ILC2s and subsequent cell survival by IFNg from type 1 lymphocytes. The confinement of ILC2s and Th2 cells to adventitia limits mortality by type 1 pathogen Listeria monocytogenes.

Summary: TGF-beta plays a critical role in allergic airway inflammation by driving effector T cells and influencing the disease outcome. Inhibiting TGF-beta can reduce the disease phenotype, decrease pro-inflammatory T cell frequencies, and increase anti-inflammatory T cell frequencies. Additionally, in patients with allergic rhinitis and asthma, allergen-specific immunotherapy can suppress Th2 and Th9 cell frequencies and decrease the secretion of TGF-beta and IL-9.

Summary: Accumulating evidence suggests that interleukin 6 (IL-6) signaling plays a crucial role in the development of allergic asthma. This study investigated the effects of IL-6 signaling and tocilizumab on TDI-induced steroid-resistant asthma. The results showed that IL-6 signaling was involved in TDI-induced airway inflammation and remodeling, and treatment with IL-6 neutralizing antibody or tocilizumab effectively alleviated these effects.

Summary: The study revealed distinct patterns of cytokine/chemokine expression in bronchoalveolar lavage (BAL) of children with severe therapy-resistant asthma, with neutrophilic asthma showing overexpression of cytokines associated with mixed T(H)17/T(H)1/T(H)2 responses, while nonneutrophilic asthma presented independently of cytokine overexpression. Neutrophilic asthma was also characterized by a higher prevalence of bacterial and viral pathogens, although cytokine expression patterns were independent of pathogen expression.

Allergen-specific immunotherapy induces the suppressive secretoglobin 1A1 in cells of the lower airways

Summary: In this study, allergen-specific immunotherapy was found to inhibit the expression of pro-inflammatory cytokines in the respiratory tract and induce the expression of other genes independently of asthma status and allergen season. In a validation cohort, an increase in SCGB1A1 was observed alongside a decrease in IL-24 during the course of AIT. SCGB1A1 was identified as a suppressor of epithelial gene expression in the airways.

Neutrophils secrete cathelicidin, which is required for maximum Th17 differentiation and plays a role in early neutrophilia directing subsequent adaptive immune responses.

Summary: This study confirms that IL-1 beta, IL-6, anti-IFN-gamma, and IL-21 promote the differentiation of dual-positive Th2-Th17 cells, and a higher proportion of these cells can be obtained through conditioned differentiation of mouse CD4(+) T cells after classical allergic asthma modeling. Prior to asthma modeling, adoptive dual-positive Th2-Th17 cells promoted differentiation of T cells into the same biphenotype cells, exacerbating asthma severity.

Through single cell RNA sequencing and adoptive transfer experiments, the researchers found that different stages of maturation and phenotype of mouse lung DC can promote Th17 or Th2 CD4(+) T cell differentiation.
