Mechanisms Underlying C-type Inactivation in Kv Channels: Lessons From Structures of Human Kv1.3 and Fly Shaker-IR Channels

Rearrangement of a unique Kv1.3 selectivity filter conformation upon binding of a drug

Summary: This study reports two structures of the human voltage-gated potassium channel Kv1.3 – apo-Kv1.3 and dalazatide-Kv1.3. Both structures are in an activated state and show remarkable rearrangements in the selectivity filter. In apo-Kv1.3, the aromatic residue Y447 adopts a unique position diverging from other K+ channels, while in dalazatide-Kv1.3, binding of dalazatide narrows the selectivity filter and stabilizes Y447 in a position similar to other K+ channels. These structural changes play a crucial role in Kv1.3's transition into the drug-blocked state.

Structural basis of gating modulation of Kv4 channel complexes

Summary: Mechanisms of modulation of Kv4 channel complexes by auxiliary subunits are central to the physiological function of channels in the brain and heart. The structures of Kv4.2-KChIP1 and Kv4.2-DPP6S complexes reveal that KChIP1 prevents N-type inactivation and stabilizes the S6 conformation, while DPP6S interacts with the S1 and S2 helices to accelerate voltage-dependent movement. KChIP1 and DPP6S do not directly interact with each other in the ternary complex, suggesting two distinct modes of modulation contribute to evoke A-type currents.

Shaker-IR K plus channel gating in heavy water: Role of structural water molecules in inactivation

Summary: In Kv channels, heavy water (D2O) slows entry into the slow inactivated state by affecting the dynamics of structural water molecules behind the selectivity filter. Increased viscosity of the external solution mimicked by glycerol has a negligible effect on the rate of inactivation. These findings support the hypothesis that the rate of structural water exchange at the region behind the selectivity filter determines the rate of slow inactivation.
