Lessons from S1P receptor targeting in multiple sclerosis

Emanuela Colombo et al.

Summary: Sphingosine 1-phosphate (S1P) is a bioactive sphingolipid that binds to specific G protein-coupled receptors expressed in various organs. Targeting S1P receptors with modulators has shown potential in treating neurological, autoimmune, and inflammatory disorders. These modulators induce lymphopenia and may have immunological effects beyond lymphocyte trafficking inhibition. They can also cross the blood-brain barrier and target CNS resident cells expressing S1P receptors. Understanding the role of S1P signaling in neuroimmunology has important implications for therapeutic approaches in other disease areas.

Endothelial S1P1 Signaling Counteracts Infarct Expansion in Ischemic Stroke

Anja Nitzsche et al.

Summary: This study explores the role of endothelial cell S1P(1) in the naive and ischemic brain as a potential target for cerebrovascular therapy. Endothelial S1P(1) plays a critical vascular protective role in the mouse brain, sustaining endothelial functions and expanding during ischemia. Disruption of this pathway exacerbates brain injury in ischemic stroke models, highlighting the importance of S1P signaling in maintaining perfusion and microvascular patency in the ischemic penumbra.

Acute Treatment With Fingolimod Does Not Confer Long-Term Benefit in a Mouse Model of Intracerebral Haemorrhage

Andrea C. Diaz Diaz et al.

Summary: This study aimed to confirm the efficacy of Fingolimod in experimental ICH using rigorous and statistically well-powered studies. The results showed no significant treatment difference in behavioral tests, contrasting with previous studies, and emphasized the importance of rigorous testing of this agent in models more representative of the clinical situation.

The effect of fingolimod on regulatory T cells in a mouse model of brain ischaemia

Kyle Malone et al.

Summary: The study found that fingolimod significantly increased the frequency of Tregs within the CD4+ T cell population in blood and spleen post-ischaemia in all three mouse cohorts. Fingolimod had a more pronounced effect on increasing Treg frequency in ischemic aged mice.

Thrombolytic strategies for ischemic stroke in the thrombectomy era

Maxime Gauberti et al.

Summary: Twenty-five years ago, intravenous thrombolysis has transformed the care for patients with acute ischemic stroke, and recent clinical trials have shown the benefits of mechanical thrombectomy. However, new strategies targeting tPA-resistant thrombi, such as tenecteplase, and experimental models targeting DNA and von Willebrand factor show promising results for improving stroke outcomes. Further research and trials are needed to enhance the effectiveness of thrombolysis in ischemic stroke patients.
