A guide to: generation and design of nanobodies

Serge Muyldermans

Summary: Nanobodies, a trademark of Ablynx, are single antigen-binding domains of heavy chain-only antibodies in Camelidae produced recombinantly for research and medical applications. Various methods are used for their generation, producing cost-effective Nbs with high affinity for their target antigens. Additionally, engineering technologies have been explored to broaden the application range of Nbs for potential advantages over other affinity reagents.

Nebulised ALX-0171 for respiratory syncytial virus lower respiratory tract infection in hospitalised children: a double-blind, randomised, placebo-controlled, phase 2b trial

Steve Cunningham et al.

Summary: This study evaluated the safety and antiviral activity of nebulised ALX-0171 in children hospitalized with RSV lower respiratory tract infection. Results showed faster viral load reduction with ALX-0171 treatment compared to placebo, but no significant improvement in clinical outcomes. Safety profile was similar between ALX-0171 and placebo groups. Future RSV antiviral studies should consider earlier intervention and more precise measurement of objective outcomes.

Structure-guided multivalent nanobodies block SARS-CoV-2 infection and suppress mutational escape

Paul-Albert Koenig et al.

Summary: In this study, new neutralizing nanobodies targeting the SARS-CoV-2 spike protein were developed, showing enhanced neutralizing activity through engineering technology and successfully suppressing the emergence of escape mutants. The findings suggest that nanobodies can neutralize through receptor binding competition and also render virions noninfectious.

Potent Neutralization of SARS-CoV-2 by Hetero-Bivalent Alpaca Nanobodies Targeting the Spike Receptor-Binding Domain

Huan Ma et al.

Summary: Cellular entry of SARS-CoV-2 relies on the binding between the receptor-binding domain of the viral Spike protein and cellular ACE2. Highly stable RBD-specific Nbs developed from immunized alpacas can block this interaction and hold promise as potential therapeutics for COVID-19.

Novalia Pishesha et al.

Summary: The COVID-19 pandemic has caused over 100 million infections and millions of deaths, prompting the development of a protein-based vaccine targeting antigen-presenting cells. This candidate vaccine shows strong humoral and cellular immunity against SARS-CoV-2 and its variants in both young and aged mice, making it a promising option for combating the virus.

Anti-CfaE nanobodies provide broad cross-protection against major pathogenic enterotoxigenic Escherichia coli strains, with implications for vaccine design

Alla Amcheslavsky et al.

Summary: ETEC is a major pathogen causing approximately 380,000 deaths globally each year, with no licensed vaccines available. Developing vaccines against ETEC is challenging due to the need for multicomponent coverage of over ten genetically variable strains.
