The assembly, regulation and function of the mitochondrial respiratory chain

Irene Vercellino et al.

Summary: The mitochondrial oxidative phosphorylation system is essential for cellular metabolism, driving ATP production through the electron transport chain. Recent studies have provided insights into the organization, assembly, and mechanisms of the respiratory complexes, including the formation of larger assemblies such as respiratory supercomplexes, and their roles in physiology.

SLC6A14 deficiency is linked to obesity, fatty liver, and metabolic syndrome but only under conditions of a high-fat diet

Sathish Sivaprakasam et al.

Summary: The 3'UTR SNP in SLC6A14 associated with obesity decreases the expression of SLC6A14, and the deficiency of SLC6A14 is linked to obesity, with this connection only evident under high-fat diet conditions.

Mitochondrial disorders of the OXPHOS system

Erika Fernandez-Vizarra et al.

Summary: Mitochondrial disorders are common inborn errors of metabolism, primarily due to dysfunction of the oxidative phosphorylation system (OXPHOS). Around half of diagnosed cases have a known genetic cause, often involving pathogenic variants in genes encoding structural subunits or factors directly involved in ETC assembly.

Perturbation of astroglial Slc38 glutamine transporters by NH4 + contributes to neurophysiologic manifestations in acute liver failure

El Hassan Hamdani et al.

Summary: The study investigates the impact of ammonia on Slc38 transporters, including competition with glutamine binding and effects on intracellular pH, membrane potential, and K+ buffering; Knocking out the Slc38a3 gene in mice results in cerebral cortical edema and disrupted neurotransmitter synthesis; A mouse model of acute liver failure shows downregulation of Slc38a3 protein, inhibited astroglial glutamine release, and cytotoxic edema.

Summary: BCAAs are crucial for skeletal muscle and whole-body anabolism and energy balance, but elevated levels and impaired metabolism may lead to chronic diseases like insulin resistance. Research is focusing on mechanisms regulating BCAA metabolism and its impact on muscle and whole-body metabolism.

Expanding Role of Proton Magnetic Resonance Spectroscopy: Timely Diagnosis and Treatment Initiation in Partial Ornithine Transcarbamylase Deficiency

Summary: A 3-year-old male patient with viral illness presented with altered mental status, emesis, and abdominal pain, leading to a quick diagnosis of ornithine transcarbamylase deficiency (OTCD) using proton magnetic resonance spectroscopy (1H MRS). Treatment with sodium phenylbutyrate and sodium benzoate initiated promptly, resulting in discharge after 3 days with no neurologic deficits. 1H MRS is a valuable tool for rapid diagnosis and treatment initiation in cases of OTCD in acute settings.
