Summary: This study investigated the impact of Klotho deficiency on arterial structure and found that it leads to severe arterial calcification and elastin fragmentation. Klotho deficiency upregulates the expression of BMP2 and RUNX2, and is independent of hyperphosphatemia in causing arterial calcification. Additionally, Klotho deficiency induces osteoblastic transition in SMCs and activates the BMP2-RUNX2 signaling pathway.
