Lipoprotein(a), LDL-cholesterol, and hypertension: predictors of the need for aortic valve replacement in familial hypercholesterolaemia

Leopoldo Perez de Isla et al.

Summary: The need for AVR due to AVS is significantly increased in FH patients, particularly in those who are older and have previous ASCVD, hypertension, increased LDL-C and Lp(a) years, and elevated Lp(a). Reducing LDL-C and Lp(a) levels, along with controlling hypertension, could slow down the progression of AVS in FH patients, but further clinical trials are needed to test this.

Genetics of 35 blood and urine biomarkers in the UK Biobank

Nasa Sinnott-Armstrong et al.

Summary: The study evaluated the genetic basis of 35 blood and urine laboratory measurements in the UK Biobank, identifying 1,857 loci associated with at least one trait. Through Mendelian randomization analysis, 51 causal relationships were discovered, including previously known agonistic effects of urate on gout and cystatin C on stroke. Finally, by developing polygenic risk scores and building 'multi-PRS' models, genetic risk stratification for common diseases was improved.

Genetic analyses identify widespread sex-differential participation bias

Nicola Pirastu et al.

Summary: Genetic analyses have revealed widespread sex-differential participation bias in population-based studies, potentially leading to incorrect inferences; the study demonstrated artifactual autosomal heritability of sex in the presence of sex-differential participation bias; through conducting a genome-wide association study contrasting one subgroup versus another, it is possible to estimate comparative biases.

Lipoprotein(a), LDL-cholesterol, and hypertension: predictors of the need for aortic valve replacement in familial hypercholesterolaemia

Leopoldo Perez de Isla et al.

Summary: Patients with familial hypercholesterolaemia (FH) have a significantly increased risk of needing aortic valve replacement (AVR) due to aortic valve stenosis (AVS), particularly in older patients with previous ASCVD, hypertension, increased LDL-C-Lp(a)-years and elevated Lp(a). Lowering LDL-C and Lp(a) levels and controlling hypertension may slow down the progression of AVS in FH, but further testing in clinical trials is needed.
