Overexpression of TaSNAC4-3D in Common Wheat (Triticum aestivum L.) Negatively Regulates Drought Tolerance

Variation in cis-regulation of a NAC transcription factor contributes to tolerance in wheat

Summary: Drought is a major limitation to wheat production worldwide, and understanding the genetic components determining drought tolerance is crucial for wheat breeding. In this study, we identified a wheat NAC gene (TaNAC071-A) that is strongly associated with drought tolerance. Overexpression of TaNAC071-A improved drought tolerance in wheat through increased expression of stress-responsive genes and improved water-use efficiency. We also found that a 108-bp insertion in the promoter region of TaNAC071-A contributes to variation in drought tolerance among wheat accessions. Importantly, introgression of this insertion allele into drought-sensitive cultivars improved their drought tolerance, making it a valuable genetic resource for wheat breeding.

The findings enriched the roles of NAC TFs in cotton drought tolerance and provided a foundation for the cultivation of transgenic cotton with higher drought tolerance.

Overexpression of the wheat NAC transcription factor TaSNAC4-3A gene confers drought tolerance in transgenic Arabidopsis

Summary: NAC transcription factors are crucial for plant responses to abiotic stresses. The study isolated three wheat TaSNAC4 homeologous genes and characterized the function of TaSNAC4-3A in plant drought tolerance. Overexpression of TaSNAC4-3A in Arabidopsis enhanced salt, osmotic and drought stress tolerance, reduced stomatal aperture size, and promoted root elongation and seed germination sensitivity to abscisic acid. Transcriptomic analysis showed upregulation of stress responsive genes in TaSNAC4-3A transgenic lines, emphasizing the important role of TaSNAC4-3A in plant drought tolerance.

Evolutionary Divergence and Biased Expression of NAC Transcription Factors in Hexaploid Bread Wheat (Triticum aestivum L.)

Summary: The study identified the evolutionary stages and expansion of NAC transcription factors in green plants, particularly in hexaploid wheat. The results showed that tandem replications during polyploidization play a significant role in increasing the number of NAC transcription factors in wheat. Additionally, the analysis of TaNACs in wheat revealed distinct groups and clusters, indicating the presence of neofunctionalization and subfunctionalization genes which are important in responding to various stresses.
