Efficacy of the NS1-truncated live attenuated influenza virus vaccine for swine against infection with viruses of major North American and European H3N2 lineages

Elien Vandoorn et al.

Summary: Control of swIAV is complicated due to multiple strains and lack of a broad cross-protective vaccine. The TX98 LAIV vaccine provides partial cross-protection against certain swIAV strains, but its efficacy against European and recent North American lineages needs further investigation.

Pathobiology of an NS1-Truncated H3N2 Swine Influenza Virus Strain in Pigs

Elien Vandoorn et al.

Summary: The study found that the H3N2 LAIV strain lvTX98 is only partly attenuated in pigs and is excreted at significant titers after intranasal vaccination. Attenuation and restricted replication of lvTX98 in vivo seemed to be associated with the loss of NS1 functions other than type I IFN antagonism.

Bat influenza vectored NS1-truncated live vaccine protects pigs against heterologous virus challenge

Jinhwa Lee et al.

Summary: Swine influenza is a significant disease for the swine industry, and traditional whole inactivated virus vaccines may cause vaccine-associated enhanced respiratory disease, while live attenuated influenza virus vaccines can effectively protect pigs from both homologous and heterologous virus infections. In this study, a chimeric bat influenza virus was used to develop a safe live vaccine to prevent swine influenza.

A Live Attenuated H9N2 Avian Influenza Vaccine Prevents the Viral Reassortment by Exchanging the HA and NS1 Packaging Signals

Sujuan Chen et al.

Summary: The H9N2 avian influenza virus is a significant zoonotic pathogen and can easily recombine with other subtypes to generate novel viruses. In this study, a new H9N2 live attenuated strain was developed to intercept reassortments and showed good genetic stability and immunogenicity in chickens.

Current and prospective control strategies of influenza A virus in swine

Hamish A. Salvesen et al.

Summary: Effective control of influenza A viruses (IAV) in pigs can be achieved through good animal husbandry practices, high animal welfare standards, and IAV surveillance programs. Vaccination, the best available tool for controlling IAV in swine, is limited by the virus's mutability and vaccine efficacy, making it far from a perfect solution. Novel genetic technologies may offer promising complementary strategies for IAV control in pigs.

Molecular Evolution of the Influenza A Virus Non-structural Protein 1 in Interspecies Transmission and Adaptation

Danyel Evseev et al.

Summary: This review discusses the species-specific molecular signatures of the influenza A virus NS1 protein in different host species and its two functions in the process of viral adaptation to new host species. The NS1 protein has the ability to broadly suppress host protein expression and inhibit antiviral interferon signaling through protein-protein interactions, which are crucial for viral replication and immune evasion. The NS1 protein demonstrates a variety of context-dependent solutions to the problem of interspecies adaptation, showing high plasticity and versatility.

Summary: The study successfully identified a specific epitope H7-12 peptide for H7N9 AIV and evaluated the protection and serological DIVA characteristics of the recombinant H7N9 AIV strain. The recombinant H7N9 AIV strain JD-cHA/17 showed good protection against H7N9 AIV challenge and could be detected using a specific peptide chip for DIVA testing.

Bat Influenza Viruses: Current Status and Perspective

Wenyu Yang et al.

Summary: Bats serve as natural reservoirs for many viruses, some of which can be transmitted to humans. Unusual influenza viruses H17N10 and H18N11 have been identified in New World bats, while an H9N2-like influenza A virus was found in Old World bats, showing characteristics similar to regular influenza A viruses. This review provides insight into the current status and future perspectives of influenza A viruses discovered in bats.

Entry of influenza A virus into host cells - recent progress and remaining challenges

Milagros Sempere Borau et al.

Summary: Influenza A viruses are a major health burden and the focus of intense research efforts, particularly in understanding the entry process into host cells for intervention strategies. Recent key findings include the identification of new entry receptors, the role of viral neuraminidase in entry, and progress in structural studies on viral hemagglutinin and matrix protein. Gaps in understanding and important questions for ongoing research efforts are also highlighted.
