African swine fever virus cysteine protease pS273R inhibits pyroptosis by noncanonically cleaving gasdermin D

Gaihong Zhao et al.

Summary: Research has found that the pyroptosis execution protein gasdermin D (GSDMD) is a new binding partner of ASFV-encoded protein S273R (pS273R). Further experiments showed that pS273R can cleave swine GSDMD, producing a shorter N-terminal fragment that does not trigger pyroptosis or inhibit ASFV replication. This discovery reveals a previously unrecognized mechanism involved in the inhibition of pyroptosis induced by ASFV infection.

Mitochondrial DNA stress triggers autophagy-dependent ferroptotic death

Changfeng Li et al.

Summary: The study reveals that zalcitabine, an antiviral drug, can suppress pancreatic cancer cell growth through inducing ferroptosis, a type of cell death. This effect is mediated by zalcitabine-induced mitochondrial DNA stress and the activation of a DNA sensing pathway, leading to cell death via lipid peroxidation. Genetically or pharmacologically inhibiting this pathway reduces the anticancer effects of zalcitabine.

Phase separation drives RNA virus-induced activation of the NLRP6 inflammasome

Chen Shen et al.

Summary: NLRP6 plays a crucial role in host defense by inducing inflammasome activation and interferon production through LLPS upon interaction with dsRNA. The poly-lysine sequence (K350-354) of NLRP6 is essential for multivalent interactions and inflammasome activation. Deficiency or mutation of Nlrp6 leads to impaired immune responses.

Human NLRP1 is a sensor for double-stranded RNA

Stefan Bauernfried et al.

Summary: Inflammasomes play a crucial role as intracellular sensors in pathogen infection and cellular perturbation. Human NLRP1 has been identified as a direct sensor for dsRNA, triggering activation during RNA virus infection. The NLRP1 complex binds dsRNA through its leucine-rich repeat domain, leading to activation through adenosine triphosphatase activity in its NACHT domain.
