Alcohol consumption and MRI markers of brain structure and function: Cohort study of 25,378 UK Biobank participants

Anya Topiwala et al.

Summary: Moderate alcohol consumption may have impacts on brain structure and function, as evidenced by associations with multiple MRI markers such as grey matter volume, white matter microstructure, and functional connectivity. These associations are stronger than those with other modifiable factors and can be exacerbated by frequent binging, high blood pressure, and BMI.

Confound modelling in UK Biobank brain imaging

Fidel Alfaro-Almagro et al.

Summary: Dealing with confounds is essential in large cohort studies to address unexplained variance and spurious correlations. The UK Biobank is a powerful resource for studying associations between imaging and non-imaging measures, but the high statistical power also increases sensitivity to confound effects. Researchers must carefully consider these factors.

Elevated iron concentration in putamen and cortical speech motor network in developmental stuttering

Gabriel J. Cler et al.

Summary: This study used quantitative mapping of brain tissue to find higher iron content in the left putamen and left hemisphere cortical regions in individuals who stutter, suggesting potential links to elevated dopamine levels or lysosomal dysfunction in stuttering. These results provide new evidence of microstructural differences in the basal ganglia and connected frontal cortical regions in individuals who stutter.

Alcohol use and dementia: new research directions

Rachel Visontay et al.

Summary: Recent evidence suggests that low-to-moderate alcohol consumption may have protective effects on dementia and cognitive function, but the methodological limitations of existing observational studies have been highlighted. Chronic heavy alcohol use can cause alcohol-related brain damage through various neurotoxic pathways, including thiamine deficiency, ethanol, and acetaldehyde. Future research should focus on robust analytical and design-based approaches to better understand the relationship between alcohol use and dementia in the general population.

Hemochromatosis Mutations, Brain Iron Imaging, and Dementia in the UK Biobank Cohort

Janice L. Atkins et al.

Summary: In individuals of European ancestry, male p.C282Y homozygotes were found to have higher brain iron deposition and a greater risk of developing dementia, while no associations were observed in female homozygotes or heterozygotes. Further studies are needed to explore whether early iron reduction interventions can prevent or slow down related brain pathologies in male HFE p.C282Y homozygotes.

A genome-wide meta-analysis yields 46 new loci associating with biomarkers of iron homeostasis

Steven Bell et al.

Summary: The meta-analysis of three genome-wide association studies revealed 62 independent sequence variants associating with iron homeostasis parameters at 56 loci, including 46 novel loci. These variants are associated with iron deficiency anemia and iron overload, highlighting their significant role in regulating iron homeostasis.
