TGF-β-mediated silencing of genomic organizer SATB1 promotes Tfh cell differentiation and formation of intra-tumoral tertiary lymphoid structures

Ricardo A. Chaurio et al.

Summary: Regulation of PD-1 expression on Tfh cells affects Tfh cell differentiation. Inhibiting SATB1 expression promotes Tfh cell differentiation and enhances immune responses. Transfer of Tfh cells can induce TLS formation and inhibit tumor growth.

Tertiary lymphoid structures generate and propagate anti-tumor antibody-producing cells in renal cell cancer

Maxime Meylan et al.

Summary: The presence of intratumoral tertiary lymphoid structures (TLS) is associated with positive clinical outcomes and responses to immunotherapy in renal cell carcinoma (RCC). B cells are enriched in TLS and demonstrate diverse maturation stages, with the presence of fully mature clonotypes. Additionally, IgG-and IgA-producing plasma cells (PCs) disseminate into the tumor beds along fibroblastic tracks in TLS+ tumors, indicating potential anti-tumor effector activity. Furthermore, the presence of IgG-stained tumor cells correlates with therapeutic responses and progression-free survival in RCC patients treated with immune checkpoint inhibitors.

IgA transcytosis and antigen recognition govern ovarian cancer immunity

Subir Biswas et al.

Summary: Most ovarian cancers are infiltrated by activated T cells that have prognostic relevance, but they show low response rates to immune checkpoint inhibitors. On the contrary, memory B cell and plasma cell infiltrates associated with better outcomes may exert protective effects against tumor cells through B cell-derived IgA in ovarian cancer.

Local induction of bladder Th1 responses to combat urinary tract infections

Jianxuan Wu et al.

Summary: Intravesical vaccination with antigens from uropathogenic Escherichia coli (UPEC) combined with a Th1-skewing adjuvant is a superior strategy to combat urinary tract infections (UTIs), especially in UTI-prone subjects. This method effectively recruits Th1 cells into the bladder to correct aberrant bladder immune responses and promote protection against UTIs.

The microbiome and human cancer

Gregory D. Sepich-Poore et al.

Summary: Microbes play significant roles in cancer, and bacteria, viruses, and fungi are believed to be key actors in cancer immunotherapy. Although the number of microbes known to directly cause carcinogenesis is small, further research is needed to understand the causal relationship between microbes and cancer.

An ex vivo tumor fragment platform to dissect response to PD-1 blockade in cancer

Paula Voabil et al.

Summary: A study using patient-derived tumor fragments as an ex vivo platform showed that the reactivation of tumor-infiltrating immune cells after PD-1 blockade could predict clinical outcomes. Tumor-resident T cells were identified as crucial in the immunological response, and a subgroup of tumors was found to be unresponsive to PD-1 blockade. Additionally, the presence of tertiary lymphoid structures in baseline tumors correlated with the capacity for intratumoral immune cell reactivation.

Summary: This study found that T follicular helper (Tfh) cells in breast cancer play a key role in the formation of tertiary lymphoid structures (TLS), with active and functional Tfh TILs in TLS being associated with positive clinical outcomes in patients. Lower Airway Dysbiosis Affects Lung Cancer Progression

Jun-Chieh J. Tsay et al.

Summary: In lung cancer patients, dysbiosis of the lower airway microbiota is more prevalent in advanced stages and associated with poor prognosis, upregulation of IL17, PI3K pathways, etc. in the airway transcriptome. The dysbiosis with V. parvula in the lower airway of the KP lung cancer model leads to decreased survival and tumor progression.
