Chemical Method to Sequence 5-Formylcytosine on RNA

This study presents a chemical method, Mal-Seq, for sequencing 5-formylcytosine (f(5)C) modifications on RNA. The study shows that high-level f(5)C modification is present in mammals but lacking in lower eukaryotes. These findings shed light on mitochondrial tRNA modifications throughout eukaryotic evolution and provide a general platform for characterizing the f(5)C epitranscriptome.

Permethylation of Ribonucleosides Provides Enhanced Mass Spectrometry Quantification of Post-Transcriptional RNA Modifications

Summary: Chemical modifications of RNA play important roles in fundamental biological processes and human diseases. However, the analysis of ribonucleoside modifications is challenging due to the hydrophilicity of the molecules. In this study, we developed a solid-phase permethylation method coupled with LC-MS/MS to efficiently analyze the modifications. We successfully identified and quantified more than 60 RNA modifications, and demonstrated the improved sensitivity, efficacy, and robustness of the new method.
