Summary: More than 20 vertebrate master sex-determining genes have been identified on different sex chromosomes of mammals, birds, frogs and fish, with some belonging to the TGF-beta signalling pathway. This pathway, despite being a subordinate member of the sex-regulatory network in mammals, has repeatedly become a top regulator, challenging current evolutionary concepts on sex-determining mechanisms.
