Tumor Microenvironment: Necroptosis Switches the Subtype of Liver Cancer While Necrosis Promotes Tumor Recurrence and Progression

Summary: Liver cancer is a diverse group of solid tumors that arise from epithelial cells and are driven by genetic and epigenetic alterations. Hepatocellular carcinoma and intrahepatic cholangiocarcinoma are the primary types of liver cancer, but the same cell type can give rise to different cancers depending on the surrounding cell death. Recent research has shown that hepatocytes can give rise to both hepatocellular carcinoma and intrahepatic cholangiocarcinoma, depending on apoptotic or necroptotic cell death of neighboring cells. The microenvironment and cell death pathway (necrosis, apoptosis, or necroptosis) play crucial roles in the development and progression of liver cancer.

Summary: This study systematically investigated the immune interaction in neuroblastoma (NBL) and identified immune-related genes with prognostic potential. The immune signature combining nine immunity genes was found to be more effective for individual risk stratification and survival outcome prediction than common clinical characteristics. A constructed nomogram based on the immune signature and clinical characteristics showed high potential for prognosis prediction.

Decreased expression of METTL14 predicts poor prognosis and construction of a prognostic signature for clear cell renal cell carcinoma

Summary: Our study demonstrated that METTL14 could serve as a favorable prognostic factor in clear cell renal cell carcinoma (ccRCC), being associated with better overall survival. Additionally, significant enrichment of METTL14 in multiple signaling pathways was identified, and a prognostic signature integrating METTL14 expression and clinicopathologic variables was established to predict survival probabilities in ccRCC. Furthermore, a METTL14-based risk score was developed with improved prognostic accuracy.

A Novel Pyroptosis-Related lncRNAs Signature for Predicting the Prognosis of Kidney Renal Clear Cell Carcinoma and Its Associations with Immunity

Summary: This research identified a predictive multi-lncRNA signature related to poor prognosis in clear cell renal cell carcinoma (ccRCC) through studying pyroptosis-related differentially expressed lncRNAs. The developed 14-lncRNA predictor showed superior performance in predicting ccRCC prognosis compared to standard clinicopathological characteristics. Additionally, immune checkpoint expressions and tumor-related pathways were found to be different between high-risk and low-risk groups in ccRCC patients, indicating potential implications for therapeutic strategies.
