EN1 Regulates Cell Growth and Proliferation in Human Glioma Cells via Hedgehog Signaling

Jinchun Chang et al.

Summary: The study found that EN1 is highly expressed in glioma cells and tissues, and its increased expression level is associated with poor prognosis. EN1 influences glioma cell proliferation, migration, and sensitivity to radiation by regulating Gli1 levels and the primary cilia-related protein TULP3. These findings suggest that EN1 plays an oncogenic role in glioblastoma pathogenesis.

The scaffolding protein DLG5 promotes glioblastoma growth by controlling Sonic Hedgehog signaling in tumor stem cells

Somanath Kundu et al.

Summary: DLG5 is upregulated in malignant gliomas and promotes invasion, viability, and self-renewal of glioblastoma stem cells. It regulates Sonic Hedgehog (Shh) signaling, making DLG5-deficient cells insensitive to Shh ligand, and its loss increases the proteasomal degradation of Gli1, leading to tumor stem cell sensitization. These findings reveal a novel and pro-tumoral role for DLG5 in glioblastoma, distinct from its proposed tumor-suppressor role in other solid tumors.

The Hallmarks of Glioblastoma: Heterogeneity, Intercellular Crosstalk and Molecular Signature of Invasiveness and Progression

Filippo Torrisi et al.

Summary: This article discusses the latest version of the World Health Organization's classification of central nervous system tumors in 2021, focusing on the biological and molecular mechanisms and tumor hallmarks of glioblastoma. The research findings indicate that the identification of glioblastoma subtypes has not led to personalized therapies and there have been no improvements in overall survival, highlighting unresolved issues. However, the evidence presented offers a promising perspective for understanding tumor pathophysiology and driving the development of new therapeutic strategies and approaches.

GANT-61 Induces Autophagy and Apoptosis in Glioblastoma Cells despite their heterogeneity

Gabriela Basile Carballo et al.

Summary: Glioblastoma is a common adult primary tumor of the CNS characterized by rapid growth and invasiveness. Resistance to chemotherapy in GBM may be due to cancer stem cells activating molecular pathways. Inhibiting the SHH signaling pathway induces apoptosis and autophagy in GBM cells, suggesting it as a potential therapeutic approach for refractory patients.

Expression of activator proteins of SHH/GLI and PI3K/Akt/mTORC1 signaling pathways in human gliomas is associated with high grade tumors

Julian Henao-Restrepo et al.

Summary: The differential expression of proteins from the PI3K/Akt/mTORC1 and SHH/GLI pathways in human gliomas suggests that activation of these signaling networks is associated with glioma progression, varying according to tumor type and grade.

The PI3K Inhibitor XH30 Enhances Response to Temozolomide in Drug-Resistant Glioblastoma via the Noncanonical Hedgehog Signaling Pathway

Ming Ji et al.

Summary: A study found that a new inhibitor XH30 demonstrates anti-tumor activity against TMZ-resistant GBM by inhibiting cell proliferation and inducing cell cycle arrest. XH30 also showed effectiveness in suppressing TMZ-resistant tumor growth in an animal model and enhancing TMZ cytotoxicity. Mechanistically, XH30 may exert its synergistic effect by repressing the key transcription factor GLI1 via the noncanonical hedgehog signaling pathway.

Primary Cilia-Related Pathways Moderate the Development and Therapy Resistance of Glioblastoma

Minghao Li et al.

Summary: Primary cilia, as microtubule-based structures, are closely related to the development of the central nervous system and play a crucial role in regulating the tumorigenesis of the central nervous system. The development of primary cilia can be regulated by several pathways, and primary cilia are also able to regulate a few signaling transduction pathways.
