Suppression of the caspase-1/GSDMD-mediated pyroptotic signaling pathway through dexamethasone alleviates corneal alkali injuries

Yuan Tan et al.

Summary: The pathological mechanism of corneal injuries mediated by alkali burns involves NLRP3-related corneal sterile inflammation. Dexamethasone can reverse the enhanced expression of pyroptosis-related proteins and inhibit pyroptosis pathway activation in corneal alkali burns.

Irisin and Secondary Osteoporosis in Humans

Roberta Zerlotin et al.

Summary: Irisin is a peptide secreted by skeletal muscle that plays an important role in bone metabolism. It promotes osteogenesis, protects osteocytes, prevents loss of bone and muscle mass, and accelerates fracture healing. Clinical evidence suggests that low levels of irisin are associated with an increased risk of osteoporosis and bone fractures. It may also serve as a marker for monitoring bone quality in secondary osteoporosis.

Itaconate inhibits ferroptosis of macrophage via Nrf2 pathways against sepsis-induced acute lung injury

Ruyuan He et al.

Summary: This study aimed to explore the regulatory role of itaconate on ferroptosis in sepsis-induced acute lung injury (ALI). The results showed that pretreatment with itaconate alleviated lung injury and inhibited LPS-induced lipid peroxidation and injury. Mechanistically, itaconate inhibited ferroptosis by regulating the accumulation and activation of Nrf2 and GPX4.

Ferroptosis: molecular mechanisms and health implications

Daolin Tang et al.

Summary: Cell death can be executed through different subroutines, such as ferroptosis which is an iron-dependent form of non-apoptotic cell death. Ferroptosis can occur through two major pathways, driven by redox imbalance and abnormal expression of redox-active enzymes. The process is precisely regulated at multiple levels, with the transcription factor NFE2L2 playing a central role in anti-ferroptotic defense.

Mycobacterium tuberculosis Exploits Focal Adhesion Kinase to Induce Necrotic Cell Death and Inhibit Reactive Oxygen Species Production

Afrakoma Afriyie-Asante et al.

Summary: Tuberculosis is a deadly respiratory disease caused by Mycobacterium tuberculosis, which manipulates host cell death to its advantage. Infection triggers a reduction in focal adhesion kinase (FAK) expression, inhibiting FAK leads to necrotic cell death, while overexpression of FAK blocks cell death completely. FAK plays an important role in host defense against Mtb infection by preventing necrotic cell death and inducing ROS production to restrict bacterial survival.

Viral dosing of influenza A infection reveals involvement of RIPK3 and FADD, but not MLKL

Teodora Oltean et al.

Summary: The study found that RIPK3 plays an important role in protection against influenza A virus, but this protection is only apparent within a limited dose range of IAV challenge. The protective outcome is independent of RIPK3 kinase activity and MLKL, suggesting that the platform function of RIPK3 is crucial for protection. FADD-dependent apoptosis also has a crucial additional effect in protection against IAV infection.

RIPK3 activation-dependent derepression of TRIM28 in cancer cells leads to enhanced immunostimulatory cytokine production and robust anti-tumor immunity.
