Summary: The study investigates the roles of chondroitin sulfate (CS) in hematopoietic cells and bone marrow niche, demonstrating that CS plays different functions in these components. The knockout of CS N-acetylgalactosaminyltransferase-1 (T1) leads to impaired repopulation of hematopoietic stem and progenitor cells (HSPCs) and activates IFN-alpha/beta signaling and endoplasmic reticulum stress. Interestingly, the T1 knockout niche is shown to better support HSPC repopulation without affecting its structure compared to the wild-type niche.
