GRB10 regulates β-cell mass by inhibiting β-cell proliferation and stimulating β-cell dedifferentiation

Summary: The levels of GRB10, a negative regulator of insulin and mTORC1 signaling, are increased in diabetic mice and high glucose-treated cells. Knockout of Grb10 in mice increased beta-cell mass and improved function, while overexpression of Grb10 induced beta-cell dedifferentiation. GRB10 is identified as a critical regulator of beta-cell dedifferentiation and mass through inhibiting mTORC1 signaling.

Vsig4+ macrophages prevent the infiltration of bacteria and their products into host tissues, but their role in the communication between microbiota and beta cells in the pathogenesis of obesity-associated islet abnormalities is unclear.
