LRRK2, GBA and their interaction in the regulation of autophagy: implications on therapeutics in Parkinson's disease

Shirley Yin-Yu Pang et al.

Summary: Mutations in LRRK2 and GBA are the most common genetic causes of Parkinson's disease, affecting the autophagic-lysosomal pathway. These genes influence each other, leading to autophagic defects, alpha-synuclein accumulation, and worsened clinical symptoms. Inhibitors of LRRK2 kinase and activators of GCase show promise in pre-clinical models, but their efficacy in idiopathic Parkinson's disease remains uncertain.

Glucocerebrosidase Activity is Reduced in Cryopreserved Parkinson's Disease Patient Monocytes and Inversely Correlates with Motor Severity

Laura P. Hughes et al.

Summary: Measuring glucocerebrosidase activity in cryopreserved peripheral blood monocytes can detect significant differences between Parkinson's disease patients and controls, with this activity inversely correlated to the severity of motor symptoms. Using cryopreserved cells can facilitate larger multi-site trials to validate monocyte GCase activity as a biomarker for Parkinson's disease.

Sex-based differences in the activation of peripheral blood monocytes in early Parkinson disease

Samantha M. Carlisle et al.

Summary: The study found significant differences in monocyte gene expression between male and female patients with Parkinson's disease, with females showing inflammatory activation and males exhibiting more heterogeneous activation patterns. These data highlight the importance of systemic monocyte activation in PD and the need for studies examining the differential effects of sex on the pathophysiology of the disease.
