Voltage compartmentalization in dendritic spines in vivo

Summary: Dendritic spines play a critical role in excitatory neurotransmission in the nervous system. The membrane potentials of spines and dendrites were measured during spontaneous activity and sensory stimulation in mice. Spines and dendrites were depolarized together during action potentials, but experienced different voltages during subthreshold and resting potentials. The regulation of voltage compartmentalization could have important implications for synaptic function and plasticity, dendritic integration, and disease states.

Summary: Corticofugal projections from the neocortex to evolutionarily ancient subcortical structures are common in mammalian sensory systems, allowing the cortex to control ascending sensory representations predictively or in a feedback manner. This study focused on the projection from the mouse auditory cortex to the inferior colliculus (IC), revealing that although individual synapses were weak, IC neurons integrated inputs from multiple corticofugal axons to generate reliable depolarizations. Descending signals reached the IC within 30 ms of sound onset and were found to nonlinearly amplify IC neurons' acoustic responses.

Summary: Activation of astrocytes and release of glutamate onto presynaptic NMDA receptors induces long-term depression in developing cortical synapses. The exact function of presynaptic NMDA receptors in this process has been debated. This study reveals that astrocyte-mediated glutamate release indirectly enhances Ca2+ influx through presynaptic voltage-dependent Ca2+ channels, contributing to the signaling cascade of long-term depression.

Dendritic branch structure compartmentalizes voltage-dependent calcium influx in cortical layer 2/3 pyramidal cells

Summary: This study reveals that back-propagating action potentials (bAPs) fail to evoke calcium influx in specific dendritic branches of mouse cortical layer 2/3 pyramidal cells, despite evoking it in sister branches. The branch-specific compartmentalization of bAP-dependent calcium signals may provide a mechanism for neurons to diversify synaptic tuning across the dendritic tree.
