Do Epstein-Barr Virus Mutations and Natural Genome Sequence Variations Contribute to Disease?

Summary: The incidence of diseases associated with EBV infection varies greatly in different parts of the world, which may be influenced by the variation in the virus genome. This article describes the main forms of EBV genome sequence variation and the potential contribution of these variations to disease development. Polymorphisms or deletions in the EBV genome can also serve as useful markers for disease monitoring. If certain strains of EBV are proven to be more pathogenic than others, immunization against these pathogenic strains may be valuable.

Summary: This study compared the genetic variations of Epstein-Barr virus (EBV) obtained from Burkitt lymphoma (BL) samples in Africa and samples from other regions. It revealed distinct genetic patterns between African and Asian EBV sequences, and identified 12 unique patterns in the LMP-1 gene. These findings have important implications for understanding the geographic and phenotypic associations of EBV variants, and for improving EBV markers of ancestry or phenotype.

Summary: This study sequenced 15 complete EBV genomes from South America and analyzed these along with 199 EBV isolates from other regions. It identified the actual contribution of each EBV gene to the geographic segregation of the entire genome for the first time. Furthermore, it established the evolution rate for the entire EBV genome based on a codivergence-independent assumption and assessed gene-by-gene evolution rates related to protein function.
