Summary: Previous studies have shown that after acute infection, foot-and-mouth disease virus (FMDV) proteins and genome can be found in the light zone of germinal centers, and this pattern is consistent with the binding of the virus to follicular dendritic cells (FDCs). This study confirmed a similar pattern of FMDV protein staining and colocalization with FDCs in mouse spleens. Blocking the binding of FMDV to complement receptor type 2 and 1 (CR2/CR1) reduced the detection of viral proteins and genomic RNA in spleen samples, as well as the neutralizing antibody titers.

Summary: Foot-and-mouth disease (FMD) is a highly contagious and economically devastating disease for cloven-hooved animals. The study identified CD4(+) T-cell epitopes in the FMD virus capsid and revealed a long-lasting Th1 cell phenotype after FMD vaccination. The presence of FMDV-specific CD4(+) T cells in bovine PBMC more than four years post-vaccination highlights the importance of vaccine-induced adaptive immunity for long-term protection.

Structures of Foot-and-mouth Disease Virus with neutralizing antibodies derived from recovered natural host reveal a mechanism for cross-serotype neutralization

Summary: This study isolated serotype O-specific neutralizing antibodies and a cross-reacting NAb from recovered natural bovine hosts, and determined virus-NAb complex structures using cryo-electron microscopy at near-atomic resolution. The findings revealed different binding modes of the antibodies to FMDV capsids, as well as a previously unknown antigenic site. These results provide valuable information for understanding FMDV neutralization and developing a universal vaccine for cross-serotype protection.

In this study, researchers identified two cross-protective antigen sites on VP2 and VP3 using bovine-derived broadly neutralizing monoclonal antibodies. One site on VP3 was recognized by bnAb C4, while the other site on VP2 was recognized by 9 different bnAbs.
